羧基-D-氨基葡萄糖对食管癌裸鼠及相关基因表达的影响

摘    要:目的观察羧基-D-氨基葡萄糖(COPADG)对食管癌裸鼠及相关基因表达的影响。方法建立食管癌裸鼠模型,随机分为对照组、COPADG组、氟尿嘧啶组(5-FU)和联合组(CF)。计算抑瘤率;用免疫组化法,检测细胞增殖核抗原(PCNA)、天冬氨酸半胱氨酸特异性蛋白酶(caspase3)、血管内皮生长因子(VEGF)表达和微血管密度(MVD);用流式细胞术,检测肿瘤细胞凋亡。结果 COPADG组、5-FU组和CF组抑瘤率分别为54.16%,38.78%,73.91%;以上3组VEGF和MVD表达均低于对照组(P<0.01),caspase3表达均高于对照组(P<0.01);COPADG组和CF组的PCNA表达低于对照组和5-FU组(P<0.01),以上3组的早期凋亡细胞率均高于对照组(P<0.01)。结论 COPADG通过抑制食管癌肿瘤细胞增殖和血管形成及诱导肿瘤细胞凋亡,抑制食管癌裸鼠肿瘤的生长。

Abstract:Objective To study the influence 2-[(3-carboxy-1-oxoprogyl) amino]-2-deoxy-D-glucose glucose(COPADG) on esophageal carcinoma nude mice,and the expression of related genes in tumors.Methods The models of esophageal carcinoma in nude mice were established.Experimental group was divided randomly into control group,COPADG group,5-FU group and combination group(CF).The rate of suppression of tumor was calculated and the expression of PCNA,caspase3,VEGF and MVD were detected by immunohistochemistry.The apoptosis of the tumors was detected by flow cytometry.Results The rate of suppression of tumor COPADG,5-FU,CF groups were 54.16%,38.78%,73.91% respectively.The expression of VEGF,MVD in COPADG,5-FU,CF groups were lower than control group(P<0.01).The expression of caspase3 in COPADG,5-FU,CF groups were higher than control group(P<0.01).The expression of PCNA in COPADG,CF groups were lower than control and 5-FU groups(P<0.01).The apoptotic cell rates in the early stage in COPADG,5-FU,CF groups were higher than control group(P<0.01).Conclusion COPADG can inhibit the growth of the human esophageal carcinoma in nude mice by suppressing proliferation,angiogenesis and induced apoptosis of the tumor.
