酸奶疙瘩的骨髓PCE微核试验

以NIH小鼠为实验动物,用食管癌高发区哈萨克族居民常用食品酸奶疙瘩,诱发其骨髓PCE的微核增高率。按浓度3.3g/ml,以10ml/kg BW腹腔或经口投药,于投药后24小时处死动物取材计数。结果提示,酸奶疙瘩的甲醇-氯仿提取物能促进NIH小鼠骨髓PCE微核率增高,与阴性对照组比较,统计上有显著意义的占测试样品的15.3％。

Micronucleus Test of Bone Marrow PCE of Mice for Hazakh''''s Cheese Cake

Micronucleus tests are known as one of the reliable in vive cytogenetic tests to detect clastobenic substances. Bone marrow polychromatic erythrocyte of NIH mice was used to estimate promotion effect of Hazakh's cheese cake on the frequency of micronuclei. The samples from the high incidence area of esophageal cancer were experimented. The animals received a methanol-chloroform extract from the samples at the concentration of 3.3g/ml dimethyl sulfoxide and 10mg/kg BW with feeding or intraperitoneally. Then they were killed by cervical dislocateon 24h after the treatment. The femoral marrow cells were flushed out with fetal bovine serum and smeared on clean slides. The smears were fixed with methanol for 5 min. and stained with Giemsa. The results show that the Hazakh's cheese cake can promote the frequency of micronuclei on bone marrow PCE of the mice. The difference was statistically significant (P<0.05 or P<0.01) in 15.3% samples.
