抗肿瘤药物敏感实验试剂盒的建立及其稳定性的观察

目的：为了对MTT法抗肿瘤药物敏感试验方法进行规范,制备成试剂盒,并对试剂盒的稳定性进行了观察。方法：试剂盒组成：（1）预加抗肿瘤药物的细胞培养板,将各种抗肿瘤药物稀释到工作浓度后加入48孔细胞培养板中,冷冻干燥,真空包装。（2）淋巴细胞分离液。（3）含10%牛血清的DMEM培养基。（4）5%的MTT溶液。（5）GKN溶液。以新鲜加抗肿瘤药物的细胞培养板为对照,测定试剂盒的稳定性。结果：由冷冻干燥、真空包装的预加药的细胞培养板组成的试剂盒保存6个月和12个月后,与对照组比较,结果没有显著性差异（P〉0.05）。结论：该试剂盒操作简便,稳定性好,利于临床推广使用。

The establishment and stability observation of the anticancer drug-sensitive test kit

Objective：To establish a kit to detect the anticancer drug sensitivity with MTT method by optimizing the experimental procedure,and the stability of the kit was observed.Methods：The kit was consists of：（1） Cell culture plate pre-coated with antitumor drugs which were diluted to work concentration,freeze-dried and vacuum-packed.（2） Lymphocyte separation medium.（3） DMEM medium containing 10% bovine serum.（4） 5% MTT solution.（5） GKN solution.Compared with plate coated with anti-tumor drugs currently,the stability of kit was determined.Results：The kit could be stored for a period and its stablity kept well.The results showed no significant difference between the two groups（P 0.05）.Conclusion：The kit is easy to use and has good stability,it might contribute to clinical application.
