摘    要:目的:探讨以发光二极管(Light-emitting diode,LED)作为光动力学疗法(photodynamic therapy,PDT)的光源,对人肺腺癌细胞株A549体外增殖的影响及其与传统大功率半导体激光器的比较。方法:采用肺腺癌A549细胞常规培养并传代,将细胞接种于底部澄清黑色96孔板内培养,加入相同浓度的血卟啉衍生物类光敏剂photosan3,分别用LED激光器(光功率为50mW)和大功率半导体激光器(光功率为200mW)采用相同光能量密度(分为三组:5J/cm^2、10 J/cm^2、20J/cm^2)进行照射。照光后用MTT法检测不同激光器PDT作用后细胞OD492值,并计算细胞存活率。结果:在光敏剂浓度相同、能量密度为5、10、20J/cm^2的条件下进行PDT,结果表明与大功率半导体激光器相比,以LED为光源对肺腺癌A549细胞进行PDT后细胞存活率更低,各组相互比较有统计学差异(P〈0.05)。结论:在相同光敏剂浓度及能量密度下采用低功率、长时间照射的LED-PDT组较半导体激光器PDT组,细胞存活率更低,LED-PDT对肺腺癌细胞株A549具有更好的杀伤效果。

Abstract:Objective:To observe the effect of light-emitting diodes(LED) as an alternative light source in photodynamic therapy(PDT) on human lung adenocarcinoma cell line A549 in vitro.To compare the different effectiveness of LED and high-power semiconductor laser on human lung adenocarcinoma cell line A549 in vitro.Methods: The lung adenocarcinoma A549 cells were resustated,conventional cultured and subcultured.The cultured lung adenocarcinoma A549 cells were inoculated at the clear bottom black 96-well plate and exposed at intensities of 5J/cm^2,10 J/cm^2,20J/cm^2 in LED laser(power 50mW)and high-power semiconductor laser(power 200mW)respectively.MTT assay was applied to measure the OD492 of each group with different light source,and then the rate of cell remnants was calculated.Results: The average cell survival rate of LED-PDT group was lower than the result of high-power semiconductor laser.Compared with semiconductor laser,the cell survival rate of LED-PDT group was lower.When under the same photosensitizer concentration,the cell survival rate was significantly different in each group(P〈0.05).Conclusion: Compared the effect of LED and semiconductor laser on lung cancer A549 cell line,the effect of LED-PDT is more powerful at the same concentration of photosensiser and laser intensity.
