血必净对梗阻肾间质炎性损害的干预实验

摘    要:目的动态观察单侧输尿管梗阻大鼠(UUO)模型肾间质炎性损害病理改变及核因子-κB(NF~κB)、转化生长因子一β1(TGF-β1)在肾间质的表达,探讨血必净对肾间质炎性损害的干预机制。方法Sprague—Dawley(简称SD)清洁级健康大鼠雌雄各半共60只,随机分为三组:假手术组(SOR组)、模型组(MOD组)、血必净治疗组(XBJ组)。每组20只;建立UUO模型,分别于手术后第7d、第14d各组随机选取10只大鼠,取梗阻侧肾组织,行HE、MASSON染色及免疫组化染色。观察肾脏间质病理变化以及NF—κBp65和TGF一β1在肾间质的表达。结果血必净能减轻UUO大鼠肾间质炎性损害(第7d、第14d肾小管间质损伤指数、肾间质胶原面积P〈0.05);血必净能下调肾组织间质NF—κBp65及TGF-β1的的表达(P〈0.05)有统计学意义。结论血必净减轻肾间质炎性损害,其机制可能通过抑制肾组织间质NF-κBp65及TGF-β1的的表达,并对早期肾间质纤维化有抑制作用。

Abstract:Objective To observe pathological changes of renal intemtital inflammation injury and the expression of NF- κBp65 and TGF-β1 in the obstructive renal tissue of UUO rats intervening by Xuebijing. Methods Sixty Sprague- Dawley rats were randomly divided into three groups: sham-operated rats group (SOR,n=20); unilateral ureteral obstructive model group (UUO,n=20),Xuebijing-treated group (XBJ,n=20). Rats in groups of UUO and SASP were all under the operation of unilateral obstruction. The ureter was dissociated but not ligated in the group SOR. Ten rats in each group were sacrificed on the 7th, 14th day after operation. The obstructive kidneys were taken out and tubulointerstitial damage was observed by HE stain and MASSON stain. The expression of NF-κB p65 and TGF-β1 in renal of mesenchyme were detected by imunohistochemistry. Results The tubulointerstitial damage in rats of UUO group was reduced after treated with Xuebijing at each time Point(at the 7th, 14th day,P〈0.05). The expression of NF- κBp65 and TGF-β1 was down-regulated in group UUO as compared to group SOR,showing signigicant difference (P〈0.01). Conclusions The kidney tubular pathological changes were aggravated along with the obstruction time. Xuebijing may reduce or prevent development of early renal interstitial injury and renal fibrosis by inhibiting the expression of NF- κB p65 and TGF-β1.
