玻璃酸钠合用姜黄素对兔膝骨关节炎的影响

目的:探讨关节内注射姜黄素加玻璃酸钠对骨关节炎软骨细胞代谢和凋亡的影响。方法:制作兔骨性关节炎模型,随机分成对照组、玻璃酸钠(SH)组、姜黄素合用(SH)组和关节炎(OA)组,每组10只。经过使用姜黄素9mg/(kg.week)合用玻璃酸钠2mg/(kg.week)和玻璃酸钠2mg/(kg.week)和生理盐水0.5ml关节腔注射治疗4周后,采用RT-PCR检测四组的Collagen-typeⅡ(CL-2)、matrix metalloproteinase-9(mmp9)、cyclo-oxygenase-2(cox-2)、Integrin-β1的基因表达情况;采用Tunel法检测软骨细胞的凋亡情况。结果:姜黄素加SH组的CL-2和Integrin-β1的基因表达均比OA组和SH组高(P0.05),cox-2和mmp9的基因表达低(P0.05)。而细胞凋亡实验也证实了这一点,软骨破坏姜黄素加SH组比OA组和SH组轻(P0.05)。结论:玻璃酸钠可以减轻软骨细胞炎症、营养软骨,而合用姜黄素可以协同玻璃酸钠减轻软骨炎症,保护软骨。

Study on Intra-articular Sodium Hyaluronate with Curcumin Solution Injections in Treating Knee Osteoarthritis of Rabbits Induced by Papain

Objective:To explore the effect of sodium hyaluronate with curcumin solution on papain induced knee joint osteoarthritis of rabbits.Methods:Established the model of osteoarthritis,rabbits were randomly divided into control group、sodium hyaluronate(SH)group 、SH with curcumin group and osteoarthritis group.Each group has ten rabbits.each group was treated by curcumin 9mg/(kg.week)with sodium hyaluronate2mg/(kg.week),sodium hyaluronate2mg/(kg.week)and Sodium Chloride 0.5ml for 4 weeks.Using RT-PCR measure the gene expression of MMP9、COX-2、Integrin-β1、 Collagen type II,Using Tunel to measure the chondrocyte apoptosis condition.Results:the gene expression of Collagen type II and integrin-β1 of SH with curcumin group is higher than osteoarthritis group and SH group,but gene expression of cox-2 and mmp9 is lower than osteoarthritis group and SH group(P<0.01).SH with curcumin group is lower than OA group and SH group in the chondrocyte apoptosis.Conclusion:SH shows beneficial effects to the papain induced knee joint osteoarthritis of rabbits and curcumin synergistically augments this effect
