蛇床子素和淫羊藿甙的提取分离及补肾壮阳作用的研究

从蛇床子和淫羊藿中分别提取蛇床子素(osthol)和淫羊藿甙(icariin),并进行补肾壮阳作用的研究。用有机溶剂提取、硅胶柱分离、波谱鉴定而得到单体osthol和icariin。用丙基硫氧嘧啶(PTU)复制“甲减”型肾阳虚模型。osthol和icariin分别给药,用放射免疫法测定小鼠血清3种甲状腺素T_3、rT_3和T_4的浓度。结果肾阳虚鼠血清T_3、rT_3和T_4浓度降低,死亡率高,而用osthol和icariin后,则有效地阻止了这3种浓度的下降,死亡率也大大降低。表明osthol和icariin能拮抗PTU的甲状腺抑制作用,预防“肾阳虚”症的出现,具有补肾壮阳作用。

STUDIES ON THE EXTRACTION AND ISOLATION OF OSTHOL AND ICARIIN AS WELL AS THIER EFFECTS OF REINFORCING KIDNEY- YANG

Osthol in Fructus Cnidii ( Shechuangzi) and icariin in Herba Epimidii (Yinyanghuo) have been extracted, isolated and identified. Their effects of reinforcing the kidney- yang and promoting virility have been studied by the authors. Propylthiouracil (PTU) was used to induce hypothyroidism in mice as model of kidney- yang insufficiency. Serum levels of thyroid hormone T_3, rT3 and T4 were determined by radioimmunoassay. The results showed that groups of the above mentioned mice treated with osthol and icariin have higher serum thyroid hormone levels and much lower mortality compared with those of the control groups, illustrated that osthol and icariin antagonize the inhibition of PTU to thyroid and prevent from kidney- yang insufficiency, and have the effects of reinforcing the kidney- yang and promoting virility.
