外阴鳞癌动物模型的建立及MDM2表达的研究

摘    要:目的:探讨外阴鳞癌动物模型建立的方法,研究在鼠外阴癌变发生、演进过程中MDM2的表达变化。方法:采用苯甲酸雌二醇注入32只129/J型鼠腹部皮下,8只为对照组,观测肿瘤的体积,取材做病理诊断。采用RT-PCR技术检测MDM2基因的mRNA表达。结果:外阴上皮内瘤变(VIN)发生率为65.5%(19/29),其中VIN1、VIN2各5例,VIN39例;外阴鳞状细胞癌(VSCC)发生率为34.5%。VIN1~2、VIN3、VSCC和对照组中,MDM2基因的mRNA的相对表达量分别为0.34±0.11、0.42±0.20、1.84±0.32和0.30±0.01;VSCC分别与VIN1~2、VIN3和对照组相比,差异均有统计学意义(P<0.05)。结论:129/J型鼠可为外阴病变(VINs/VSCC)的研究提供动物模型。MDM2在鼠外阴鳞癌中的变化与人组织相同,MDM2可作为外阴癌变的生物学指标。

Abstract:Objective:To study the way of establishing an animal model for the development of vulvar squamous cell carcinomas and analyze the expression of MDM2 mRNA.Methods:Estradiol benzoate hypodermically was injected to the abdomen of 32 129/J mice on the dose of 15mg/kg,no treatment to control group.Samples were observed under the light microscope.The expression of MDM2 mRNA was detected with RT-PCR.Results:VIN of mice was found in slice that stained by HE method,the incidence of VIN was 65.5%,there were 5 cases of VIN1,5 cases of VIN2,and 9 cases of VIN3.The incidence of VSCC was 34.5%.MDM2 mRNA was highly expressed in the VIN1-2,VIN3 and VSCC tissue,the expression of MDM2 had not been found in the vulvar epithelium of the comparison mice,there was no statistical significance between the comparison group and VINs group(P>0.05).there was statistical significance between comparison group and VSCC group or between VINs group and VSCC group(P<0.05).Conclusion:Formation a VINs/VSCC model of mice successfully by using artificial estrogen.The change of MDM2 in VSCC of mice is the same as which in human tissue,so we can use MDM2 as a biological marker of VSCC.
