郑州地区汉族正常人群、糖尿病和糖尿病并发冠心病患者芳香酯酶Q192R基因多态性

摘    要:目的:研究郑州地区汉族正常人群、2型糖尿病(DM)及DM并发冠心病(DM-CAD)患者芳香酯酶(Arylesterase/Paraoxonase,ArE/PON1)192位基因多态性。方法:用比色法测定郑州地区汉族正常人群(61例)、DM(60例)和DM-CAD(67例)患者血清Ar/PON1活性,用聚合酶链式反应分析技术检测ArE/PON1基因192位多态性,用酶法和酶联免疫法分别测定血清胆固醇、高密度脂蛋白胆固醇(HDLc)、氧化型低密度脂蛋白(oxLDL)等指标。结果:郑州地区汉族人群中存在有ArE/PON1192位基因多态性,Q、R基因频率健康人群中分别为0.56与0.44;DM组为0.53与0.47;DM-CAD组为0.38和0.62。DM-CAD组R基因频率高于DM组和对照组。基因型百分比DM-CAD组RR(31%)高于DM组(13%)和对照组(11%)(P<0.05)。DM与DM-CAD组血清ArE/PON1活性水平低于对照组,以DM-CAD组为最低(P<0.05)。酶活性的降低与HDLc、HDL2C呈正相关,与oxLDL呈负相关。结论:郑州地区汉族人群存在ArE/PON1 192位基因多态性,并提示R基因是郑州地区汉族人群DM-CAD的危险因素。

Abstract:Aim:To investigate the relationship between arylesterase/paraoxonase(ArE/PON1) Q192R gene polymorphism and type 2 diabetes mellitus (type 2 DM) complicated with coronary artery disease (CAD) in population of Han nationality in Zhengzhou area.Methods:Serum ArE/PON1 activity and its Q192R gene polymorphism were determined in 60 patients with type 2 DM, 67 DM patients complicated with coronary artery disease(DM CAD),and 61 healthy subjects. ArE/PON1Q192R gene polymorphism were detected by PCR method.Results:The frequencies of the Q allele and R were 0.56 and 0.44 in healthy subjects,0.53 and 0.47 in patients with DM,0.38 and 0.62 in patients with DM CAD,respectively. The R gene frequency in DM CAD group was higher than that in DM group and healthy subjects. The percentage of RR genotype in DM CAD group (31%)was higher than that (13%) in DM group and that (11%)in healthy subjects.Serum ArE/PON1 activity in DM and DM CAD groups were lower than that in healthy subjects and it was the lowest in DM CAD group.There were a positive correlation between ArE/PON1 activity and HDLc,HDL2C,and a negative correlation between ArE/PON1 activity and oxLDL.Conclusion:ArE/PON1Q192R polymorphism exists in Zhengzhou population of Han nationality. It is suggested that R allele might be a risk factor for DM complicated with CAD in Zhengzhou population of Han nationality.
