冠心平对H2O2致血管内皮细胞损伤的影响

摘    要:目的:观察冠心平对H2O2致血管内皮细胞损伤过程中NOS与ET的影响。探讨冠心平治疗冠心病的可能机制。方法:体外培养内皮细胞ECV304,用100umol/L H2O2损伤细胞,随机分成正常对照组、模型对照组、阳性药对照组、冠心平大、中、小剂量组。用血清药理学方法给药,继续培养后显微镜下观察细胞生长情况,检测NOS与ET。结果:100umol/L H2O2对ECV304有损伤作用。冠心平能显著提高NOS的活性;显著减少H202诱导ECV304ET的产生。结论:冠心平可降低H2O2对ECV304的损伤程度,其机理可能与其提高NOS活性,增加NO合成,并且减少ET的合成,从而调节血管舒缩功能有关。

Abstract:Objective: To observe the effect of Guanxinping on H2O2 induced coronary vascular endothelial cells injury and to Investigate the possible mechanism that Guanxinping cures coronary heart disease.Methods: Cultured endothelial cells ECV304,with 100umol / L H2O2 damage cells,were randomly divided into normal control group,model group,positive control group,low dose group,middle dose group,high dose group.With serum pharmacological administration,cultured cell growth was observed under the microscope and NOS and ET were detected.Results: 100umol / L H2O2 can injure ECV304.Guanxinping significantly increased NOS secretionand significantly inhibited the level of ET.Conclusion: Guanxinping can reduce the damage of ECV304 induced by H2O2,the mechanism may be related to antioxidant effects.
