附子甘草配伍减毒增效/存效实验研究

目的观察附子配伍不同比例的甘草对附子毒性和药效的影响。方法 ICR小鼠,0.4 mL/10 g体质量灌胃给药,平行比较附子和附子配伍不同比例甘草的急性毒性,测定其半数致死剂量（LD50）;SD大鼠2 mL/100 g体质量十二指肠注射给药,平行比较附子和附子配伍甘草的心脏毒性,测定其半数中毒剂量（TD50）;采用高效液相色谱法测定附子和附子与甘草配伍后水煎液附子中毒性成分;通过抗炎和镇痛实验观察附子配伍甘草对附子药效的影响。结果随着甘草配伍比例的增高,附子急性毒性LD50和附子心脏毒性TD50增大,附子配伍甘草后合煎液毒性成分低于单煎液,但附子配伍甘草后对附子抗炎和镇痛药效没有明显影响。结论甘草配伍附子在减轻附子毒性的同时不影响附子的药效,甘草对于附子毒性具有剂量依赖性拮抗作用,附子配伍甘草减毒时应该考虑甘草的配伍比例。

Experimental Study on Effects of Radix Glycyrrhizae on Toxicity and Efficacy of Radix Aconiti Carmichaeli in Compatibility

Objective To study effects of Radix Glycyrrhizae on the toxicity and the efficacy of Radix aconiti carmichaeli in a combination with different ratio between them.Methods Acute toxicity,heart toxicity and toxic ingredients of Radix aconiti carmichaeli were comparatively measured when it was used in a combination with different doses of the liquorice.At meantime,the effects of the liquorice on anti-inflammatory and analgesic activities of Radix aconiti carmichaeli were observed in the drug combination.Results With increase of the liquorice dose in the combination,both LD50 value in acute toxicity and TD50 value in heart toxicity of Radix aconiti carmichaeli were positively increased,while level of the toxic components in the water-extract of the combination of Radix aconiti carmichaeli and the liquorice was lower than those in the water-extract of Radix aconiti carmichaeli alone.On the other hand,the anti-inflammatory and analgesic effects of Radix aconiti carmichaeli were not influenced by liquorice in the combination.Conclusion The liquorice appeared to antagonize the toxicity of Radix aconiti carmichaeli in a pattern of dose-dependency,but it cannot change therapeutic effect of Radix aconiti carmichaeli.When liquorice and Radix aconiti carmichaeli were decocted together,liquorice was able to decrease the level of toxic aconitines in a dose dependent manner.The ratio or the dose of liquorice should be considered when it is combined with Radix aconiti carmichaeli for detoxification.
