RP-HPLC测定腰痛合剂中次乌头碱和乌头碱的含量

目的建立腰痛合剂中次乌头碱和乌头碱的含量测定方法。方法采用反相高效液相色谱法测定腰痛合剂中次乌头碱和乌头碱含量,色谱柱SunFire C18（4.6 mm×250 mm,5μm）,流动相为乙腈-0.2%冰乙酸（用三乙胺调pH为6.30）,梯度洗脱,流速1 mL/min,检测波长232 nm,柱温为30℃。结果次乌头碱和乌头碱分别在进样量0.0072～0.072μg、0.004 6～0.046μg范围内与峰面积呈良好的线性关系,r分别为0.999 5、0.999 8,平均加样回收率分别为98.2%（RSD=1.6%）、97.4%（RSD=1.9%）。结论该方法准确、方便、快速,可以用来测定腰痛合剂中次乌头碱和乌头碱的含量。

Determination of Mesaconitine and Aconitine in Yaotong Mixture by RP-HPLC

Objective To develop a method for detetmining mesaconitine and aconitine in Yaotong Mixture.Methods RP-HPLC was used for the determination of mesaconitine and aconitine.The column was Sunfire C18(4.6 mm 250 mm,5 μm).The mobile phase was acetonitrile-0.2% acetic acid(adjusted pH value at 6.30 by triethylamine),and the gradient elution was used.The detection wavelength was set at 232 nm,and the flow rate was 1.0 mL/min.Results Mesaconitine and aconitine showed a good linear relationship in the range of 0.007 2-0.072 μg(r=0.999 5) and 0.004 6-0.046 μg(r=0.999 8) respectively.The average recovery of mesaconitine was 98.2% with RSD of 1.6%(n=6),and the average recovery of aconitine was 97.4 % with RSD of 1.9%(n=6).Conclusion The method is accurate,simple and fast,and can be used for the determination of mesacontine and aconitine in Yaotong Mixture.
