缺血-再灌注对股骨头关节软骨细胞分化及基质稳定性的影响

摘    要:目的:探讨缺血-再灌注对股骨头关节软骨退行性变的影响。方法:建立髋关节缺血再灌注模型,大鼠随机分为正常组、模型组。于术后不同时点取股骨头关节软骨进行苏木精-伊红染色观察缺血-再灌注后股骨头关节软骨的病理组织学结构改变;利用Van Gieson胶原纤维染色法观察胶原纤维沉积变化;观察VEGFmRNA原位杂交表达。结果:关节软骨形态结构逐渐紊乱,再灌注后软骨细胞明显减少,差异均有显著性意义(P﹤0.01)。结论:缺血-再灌注可加重股骨头关节软骨的损伤,细胞增殖活性下降及细胞环境不稳定可能是缺血-再灌注早期股骨头关节软骨损伤的重要机制。

Abstract:Objective:TO study effect of ischemia-reperfusion by articular cartilage degeneration upon femoral-head.Methods: Establish the hip joint of is chemia reperfusion model the rats.The rats were random divided into the normal groups and the model groups.In the postoperative time points From the articular cartilage of femoral head,Observe the structure of articular cartilage including cartilage cells,the change of collagen and the expression of VEGFmRNA by HE,Van Gieson,and in Situ Hybridization.Results: The model rats' articular cartilage developed with the course of illness,disordered structure,the cartilage cells became few and.small,fiber rose.The expression of VEGFmRNA reduced,the microvessel density enlarged.Conclusion: With the extension of the course,Ischemia reperfusion can exacerbate of articular cartilage of femoral head injury,Cell apoptosis and environmental instability may be ischemia reperfusion of early femoral head articular cartilage injury mechanism.
