不同转移潜能的人体横纹肌肉瘤细胞系细胞骨架的研究

摘    要:目的 研究细胞骨架与三种人体不同转移潜能横纹肌肉瘤细胞系的关系。方法 采用直接和间接免疫荧光法 ,用激光扫描共聚焦显微镜观察细胞骨架F actin、vinculin、tubulin、vimentin及desmin的结构及分布 ,同时测定各种细胞骨架的含量。结果 横纹肌肉瘤细胞微丝和微管骨架均有数量减少及发育不良 ,其完善程度与横纹肌肉瘤的转移潜能呈负相关 (P <0 .0 5 ) ;F 肌动蛋白小体的出现频率与横纹肌肉瘤的转移潜能呈正相关 (P <0 .0 5 ) ;中间丝在横纹肌肉瘤中结构无明显异常 ,荧光强度两两比较差异无显著性 (P >0 .0 5 )。结论 不同程度细胞骨架的异常可能与横纹肌肉瘤的转移潜能不同有关 ,其有可能成为判断横纹肌肉瘤恶性程度及预后的指标之一。

Abstract:Objective To investigate the relationship between cytoskeleton and metastatic potential in three human rhabdomyosarcoma (RMS) cell lines. Methods Direct and indirect immunofluorescent stainings were used to detect and to quantify RMS cells' cytoskeletons, such as F actin, vinculin, tubulin, vimentin and desmin, in compare with cultured normal human myoblasts under laser scanning cofocal microscope. Results Immunofluorescent staining of cytoskeletons showed that microfilament cables (stress fibers) and microtubules were reduced dramatically in number and were maldeveloped in three cell lines of RMS. There was a negative correlation between the well formation of cytoskeletons and metastatic potential in RMS cell lines( P <0.05).The frequency of F actin aggregates increased as metastatic potential increased( P <0.05). There were little structure disruption of intermediate filament, which included vimentin and desmin in RMS cell lines, compared with cultured myoblasts. The fluorescent intensity between RMS and myoblasts had no significant difference ( P >0.05). Conclusion Maldevelopment of cytoskeletons in varying degrees may correlate with different metastatic potential of RMS. It may help to determine the malignant behavior of RMS and become a prognostic index of RMS.
