胚胎大鼠神经干细胞的培养及鉴定

摘    要:目的:探讨胚胎大鼠神经干细胞培养,传及鉴定方法。方法:从胚胎大鼠脑室下区(SVZ)组织分离得到神经干细胞,采用无血清培养技术进行培养,并诱导分化,采用SABC法对分化后后的细胞进行神经元特异性烯醇化酶(NSE)、胶质纤维酸蛋白(GFAP)检测及姬姆萨染色进行细胞鉴定。结果:成功培养出了胚胎大鼠神经干细胞,培养出的细胞具有自我更新能力的增殖能力,可分化为神经元,神经胶质细胞及少突胶质细胞,结论:胚胎大鼠神经干细胞在体外适宜的培养条件下具有自我更新,增殖和多向分化潜能。

Abstract:Aim:To incubate,proliferate,and identify the neural stem cells originated from embryonic rat brains. Methods ;The cells isolated from embryonic rat brains were incubated with serum-free medium. The capacity of proliferation and the characteristics of differentiation of the neural stem cells were tested with inlmunocytochemistry. Results: The cells of embryonic rat brains were able to self-renew and proliferate in serum-free medium, and their differentiated offsprings expressed the specific antigens of neurons and glial cells respectively. Conclusion; The neural sphere cells, which were incubated in uitro are able to self-renew,proliferate,and are multipotent. So,they are neural stem cells.
