六味地黄和金匮肾气丸对实验性自身免疫性脑脊髓炎小鼠淋巴细胞亚群和NK细胞的影响

目的:观察六味地黄丸和金匮肾气丸对实验性自身免疫性脑脊髓炎(EAE)小鼠淋巴细胞亚群和自然杀伤(NK)细胞的影响。方法:皮下注射髓鞘少突胶质细胞糖蛋白_35-55(MOG_35-55)与完全弗氏佐剂(CFA)混合的抗原,辅以腹腔注射百日咳毒素(PTX),建立EAE模型。每天记录各组小鼠体重及神经功能评分;造模后第12,25及40天,用流式细胞仪检测小鼠外周血淋巴细胞亚群及NK细胞;取各组小鼠脑和脊髓,进行病理观察。结果:模型组小鼠造模后第9天开始陆续发病,第22天达到高峰,神经功能评分为4.0分,体重明显降低,淋巴细胞亚群中CD8⁺明显降低,CD4⁺/CD8⁺比值明显升高,NK细胞水平明显降低。各治疗组小鼠的神经功能评分均比模型组降低,病程缩短,CD8⁺,CD4⁺/CD8⁺比值及NK细胞水平基本调节至正常。病理观察发现模型组小鼠大脑和脊髓病变表现为小静脉周围炎性细胞浸润形成袖套样改变及神经核固缩。激素组、六味地黄丸组和金匮肾气丸组均有不同程度减轻,病灶区域的炎性浸润细胞数较少,袖套样改变少见。结论:六味地黄丸和金匮肾气丸均有防治EAE的作用。其作用机制可能与调节细胞免疫系统有关。六味地黄丸在改善EAE小鼠体重、神经评分及调节细胞免疫等方面要优于醋酸泼尼松和金匮肾气丸。

The Effects of Liuweidihuang Pill and Jinguishenqi Pill on Lymphocyte Subgroups and Natural Killer Cells in Mice with Experimental Autoimmune Encephalomyelitis

Objective:To observe the effects of Liuweidihuang Pill and Jinguishenqi Pill on lymphocyte subgroups and natural killer(NK) cells in mice with experimental autoimmune encephalomyelitis(EAE).Methods:The mice were immunized subcutaneouly with MOG_35-55 emulsified in complete Freund's adjuvant(CFA) and intraperitoneally Pertussis toxin(PTX) for establishing EAE model. The animals were weighted and graded daily for clinical disability.At 12,25 and 40 days after immunization,lymphocyte subgroups and NK cells in the peripheral blood of mice were detected by flow cytometer,brain and spinal cord of the mice were harvested and the pathological changes were studied.Results:The mice of model group onseted in succession at day 9.The clinical symptoms climaxed at day 22 with a highest score of 4.0.The weights of the mice were lowed obviously.CD8⁺ in the lymphocyte subgroups and NK cells were decreased obviously,the value of CD4⁺/CD8⁺ was increased obviously.Clinical score of remedial group reduced and the courses of disease were shortened.The lymphocyte subgroups and NK cells were adjusted to normal state after treated by the drugs.Pathologi-cally,infiltrated inflammatory cells were observed around vein to form sleeve,the nuclei were pyknosis in model group mice.The inflammatory cells foci comparatively decreased,sleeve-like changes were seldom seen in prednisone,in Liuweidihuang Pill and Jinguishenqi Pill treated groups.Conclusions:Liuweidihuang Pill and Jinguishenqi Pill have the effects on prevention and treatment the mice with EAE.The mechanism may regulate the cell immune system.Liuweidihuang Pill is superior to prednisone acetate and Jinguishenqi Pill in improving the weight of EAE mice,nerve score and regulating the immune cells.
