竹叶提取物对犬实验性心肌梗死的保护作用

摘    要:目的研究竹叶提取物对犬实验性心肌梗死的保护作用 ,以便为竹叶提取物的临床新用途提供实验依据。方法采用麻醉开胸犬 ,通过结扎左冠状动脉前降支 (LAD)产生急性心肌梗死模型 ,从心肌梗死面积 (MIS) ,心肌梗死程度及血清酶学变化观察竹叶提取物的抗心肌缺血作用 ,并从心肌氧代谢及自由基损伤等角度分析其可能的作用机制。结果竹叶提取物 (5、10、2 0mg·kg-1)可降低血清肌酸磷酸激酶 (CK)、乳酸脱氢酶 (LDH)、天冬氨酸转移酶 (AST)活性 ,并能提高超氧化物歧化酶 (SOD)活性 ,显著降低心肌梗死面积。结论竹叶提取物对急性缺血心肌产生明显保护作用 ,作用机制可能与其对抗自由基损伤 ,增强体内抗氧化酶活性 ,减少心肌耗氧量 ,减少心肌梗死面积有关。

Abstract:Objective To study the protective effects of bamboo leaves extract(BLE)on experimental dog myocardial infarction.Methods Models of acute myocardial infarction were prepared by ligating the left anterior descending coronary artery of a dog.The anti-myocardial ischemia action was assessed by determing the changes in myocardial infarct size (MIS),(-ST and N-ST) and serokinases.Its probable mechanism of action was proposed in view of the resultant myocardial metabolic changes and free radical injury.Results The activities of serum creatine phosphokinase and lactic dehydrogenase were also decreased,and the activities of superoxide dismutase were enhanced.At the same time,BLE (5,10, 20 mg·kg~ -1) also decreased the MIS in dogs induced by coronary occlusion.Myocardial oxygen consumption was also lowered.Conclusion These findings suggest that BLE is beneficial for myocardial ischemia,probably by inhibiting oxygen free radical formation and enhancing endogenous antioxidase activity,reducing myocardial oxygen consumption and reducing the MIS in dogs.
