摘    要:目的 探讨结核分枝杆菌(结核菌)对链霉素(Sm)产生耐药性的分子机制,建立直接快速检测结核菌Sm药物敏感性的试验方法。方法 应用聚合酶链反应-单链构象多态性(PCR-SSCP)银染分析技术检测32例结核菌临床分离株的rpsL基因。结果 以结核菌标准株(H37Rv)为对照,32株结核菌临床分离株中,10株Sm敏感株的rpsL基因未见SSCP泳动异常,22株Sm耐药株中,16株(72.7%)的rpsL

Abstract:Objectives To study the molecular mechanism of M. tuberculosis resistance to streptomycin, and to set up a method for direct, rapid detection of streptomycin resistance mutations in M. tuberculosis. Method The rpsL genes of 32 M. tuberculosis clinical isolates were tested by PCR SSCP. Results Strain H 37 R v was used as a control. In 32 M. tuberculosis clinical isolates, the rpsL PCR fragments form 10 drug susceptible isolates had no differences in rpsL PCR profiles with strain H 37 R v. In 22 streptomycin resistant isolates,16 showed apparent differences in SSCP profiles.Conclusions The rpsL gene mutation is an important mechanism of M. tuberculosis resistance to streptomycin. PCR SSCP might be used as a simple, rapid and reliable diagnostic method for M. tuberculosis drug resistance.
