电镜快速诊断婴儿巨细胞病毒感染

本文报告以琼脂糖凹杯与普通滤纸组成的吸水系统浓集尿标本,以电镜快速诊断婴儿 HCMV 感染。在129例无症状和一般疾患婴儿尿液中,23例检出 HCMV 颗粒。有包膜的颗粒直径为170～185nm,核衣壳直径约100nm。在核衣壳上有排列规则的壳微体。尿液中所含的病毒颗粒在人胚肺成纤维细胞中引起胞核和胞浆内包涵体的特征性改变、鉴定为 HCMV。初步观察表明,应用电镜快速诊断婴儿 HCMV 感染是可行的。

Rapid Detection of Cytomegalovirus Infection in Infants by Electron Microscopy

This paper described the rapid detection of human cytomegalovirus (HCMV)infection in infant by direct electron microscopy(DEM).The urine samples obtained from 129 cases of asymptomatic and symptomatic infants were concentrated by the adsorb-water system,which was compo- sed of agarose and filter paper.Viral particles found in 23 cases out of 129 infants were exhibited morphologically herpesvirus characteristics: Enveloped virion had 170 to 185nm in diameter and the capsid had approximately 100nm in diameter,Capsomeres were ranged regularly on the outer of core.The herpesvirus detected by DEM were demonstrated by typical cytopathological effects and intranuclear inclusions in cell culture methods to be HCMV.The preliminary observation suggests that it might be practical to detect rapidly HCMV in urine specimens by DEM.
