抑癌基因TESTIN在子宫内膜癌组织中的表达

摘    要:目的探讨TESTIN基因在子宫内膜癌发生、发展中的作用,为子宫内膜癌诊治提供依据。方法采用qRT—PCR法和免疫组织化学法检测68例子宫内膜癌组织中TESTIN基因的表达,分析其表达与临床病理特征的关系。结果76.5%(52/68)的子宫内膜癌组织中TESTIN蛋白表达为阴性或弱阳性,而癌旁正常的子宫内膜组织中TESTIN表达均为阳性或强阳性(x2=18.133,P〈0.01),子宫内膜癌组织中TESTINmRNA表达水平(0.61±0.23)明显低于癌旁正常内膜组织(1.26±0.18,t=6.500,P〈0.01)。子宫内膜样腺癌组TESTIN表达下降率较子宫乳头状浆液性腺癌组明显升高,差异有统计学意义(X2=10.35,P=0.001)。但年龄、病理分化程度、病理分期、组织浸润及有无淋巴结转移与TESTIN表达下降均无关(P〉0.05)。结论TESTIN基因在子宫内膜腺癌中发挥抑癌功能,其表达下降甚至缺失是子宫内膜癌发生的重要因素之一。

Abstract:Objective To investigate the expression of TESTIN gene in endometrial carcinoma and explore the functions of this gene in tumor development and progression.Methods qRT-PCR and immunochemical staining assay were used to determine the mRNA and protein level of TESTIN in the tumor tissues,and the relationship between TESTIN expression and clinical pathology characteristics was analyzed.Results Compared to normal tissue,76.5% (52/68) tumor tissues showed TESTIN reduced ( P < 0.01 ),furthermore,this reduction in the subgroup of endometrioid adenocarcinoma was significant,but it was rarely observed in the subgroup of serous papillary adenocarcinoma.Conclusions TESTIN was obviously down regulated in endometrail carcinoma,especially in endometrioid adenocarcinoma,which indicated TESTIN played an important role in tumorigenesis of uterine.
