骨髓间充质干细胞治疗再生障碍性贫血的研究

摘    要:目的探讨骨髓间充质干细胞(mesenchymal stem cells,MSC)治疗再生障碍性贫血(aplastic anemia,AA)模型鼠的可行性。方法建立免疫介导AA小鼠模型。输入体外增殖培养的BALB/c小鼠骨髓MSC,作为MSC组,15只;用未治疗的AA模型小鼠作为第一对照组(模型组),15只;正常BALB/c小鼠作为第二对照组(正常组),10只。把MSC输注到实验组小鼠体内,分别于第1周、第2周、第3周、第4周观察各组小鼠的存活率、血常规、骨髓形态学变化等各项指标。结果模型组小鼠,3只于第2周内死亡,7只于第3周内死亡,2只于第4周内死亡,28d存活率20.0%;MSC组,4只于第3周内死亡,28d存活率73.3%,与模型组比较差异有统计学意义(P〈0.05)。AA小鼠模型组与MSC组在8d时外周血象明显下降,14d时模型组外周血象仍明显降低,而MSC组开始回升,至28d血象基本恢复正常。骨髓行形态学检查显示模型组骨髓增生明显减低,有核细胞数明显减少,由大量脂肪细胞填充骨髓,而MSC组骨髓增生活跃,脂肪细胞较少。结论骨髓间充质干细胞可以提高再生障碍性贫血小鼠存活率,改善骨髓造血功能,对再生障碍性贫血模型鼠有治疗作用。

Empirical Study of Bone Marrow Mesenchymal Stem Cell Treating Aplastic Anemia

Abstract:Objective To explore the feasibility of bone marrow mesenehymal stem cells (MSC) treating model mice with aplastic anemia(AA). Methods Immuno mediated aplastic anemia mice model was established according to Yao' s method. Forty BALB/c mice were divided into AA model group( n = 15 ) , MSC group( n = 15 ) and normal group( n = 10). In MSC group, MSC of enrichment culture in vitro was infused by intravenous injection at 5 days after establishment of model. Survival rate of mice, transmutation of blood count and morphology features of bone marrow were observed in all groups after 1,2,3 and 4 weeks. Results The survival rate of mice in MSC group was 73.3% which was higher than that of group in AA model (20.0%, P 〈 0.05 ). Pancytopenia was found in model group and MSC groups at day 8, the number of white blood cells in peripheral blood of AA model group at day 14 after establishment of model was significantly lower than those in MSC group and number group, respectively, while at day 28 the pancytopenia recovered nearly normal level in MSC groups. In MSC group bone marrow hyperplasia and the number of nucleated cells were significantly higher than those in AA model group. Conclusion MSC infusion can reduce the degree of bone marrow failure and improve the bone marrow hematopiesis function and survival of immuno mediated AA mice model.
