| 摘 要： | 目的构建卵巢特异启动子-1（ovarian-specific promoter,OSP-1）调控白介素-24基因（Interleukin-24,IL-24）表达的真核表达载体。方法通过RT-PCR方法从人外周血基因组中扩增IL-24基因片段,应用基因重组技术将0SP-1启动子片段插入到pcDNA3.0真核表达载体,替换pcDNA3.0载体的CMV启动子与增强子序列,然后将克隆的IL-24基因片段插入到pcDNA3.0-OSP-1载体的OSP-1启动子的下游,从而构建成由OSP-1启动子调控IL-24基因表达的载体pcD-NA3.0-OSP-1-IL-24。结果载体构建过程的酶切结果以及测序结果表明载体pcDNA3.0-OSP-1-IL-24构建正确。结论通过基因重组技术成功构建了卵巢特异启动子调控IL-24基因表达的真核表达载体,为体外研究IL-24靶向特异性杀伤卵巢癌细胞提供了前期实验基础。 |
