摘    要:目的 观察IL-2真核表达载体(pWRG3169)对HBV-SDNA疫苗(pCR3.1-S)诱导Balb/c小鼠(H-2^d)的特异性细胞免疫应答及其对稳定表达HBsAg的小鼠肥大细胞瘤P815细胞(P815-HBV-S)成瘤性的影响。方法肌肉注射DNA疫苗,背部皮下接种P815-HBV-S细胞,观察成瘤情况,4h^51Cr释放法检测小鼠脾细胞CTL活性。结果 对照组、pCR3.1-S及共同免疫

Abstract:Objective To observe the effect of interlekin 12 eukaryotic expression plasmid on specific cellular immune responses and the protection against P815 mastocytoma Cells stably expressing HBV surface antigen after immunized with HBV gene vaccine in mice. Methods The immunization was performed by intramuscular injection in this expecriment. Three weeks later, P815- HBV- S was directly inoculated into mice by subcutaneous injection. The tumor growth was measured every five days and HBsAg speiFic cytotoxic T lymphocytes (CTLs) activity was measured by 51 clromium release assay. Results The HBV - S gene vaccine evidently inhibited the tumor growth, prolonged the survival peroid(> 38.2 d) and improved the survival rate in mice, which was promoted by eukaryotic expression plasmid of interleulkin 12. Meanwhile, the HBsAg specific CTLs activity induced by PCR 3. 1 - S was Obviously increased after codelivered IL - 12 vaccine. Conclusion pCR 3. 1 - S gene vaccine has a strong antigenicity in cellular immunity and had marked killing effect of HBV infected cells in vivo. Cene vaccine against HBV may be usefrl for both prophylactic and therapeutic purposes. This effect can be promoted by interleukin 12 eukaryotic expression plasmid.
