急性巨核细胞白血病癌基因表达及其意义研究

摘    要:采用生物素标记探针核酸分子原位杂交(ISH)对5例急性巨核细胞白血病(acutemegakaryoblastic leukemia,AMKL,FAB-M7)患者细胞的癌基因表达进行了观察。2例慢性粒细胞白血病(CML)巨核细胞变患者的N-ras、c-sis、c-abl和c-myc mRNA高表达,其中1例同时高表达c-fms;1例骨髓增生异常综合征(MDS)转化M7患者c-mvc、c-sis和c-fms mRNA高表达;另2例急性非淋巴细胞白血病-M7(ANLL-M7)患者c-myc和c-fms mRNA高表达。本文对癌基因与AMKL恶性克隆扩增和急性非淋巴细胞白血病巨核细胞多倍体化障碍关系进行了讨论。

Abstract:Acute megakaryoblastic leukemia (AMKL, FAB-M7) is considered a rare human malignant hematologic disorder. It is characterized by dense medullary overgrowth of reticulin and by bone marrovv infiltration of small mononuclear megakaryoblasts. However, the mecha-nism of megakaryoblast proliferation is stili poorly understood. In this report, the exprssion of N-ras, c-myc, c-sis, c-abl and c-fms oncogenes of bone marrovv and peripheral blood mononuclear celis from five AMKL patients was investigated by in situ hybridization technique. The results shovved that the mRNA expressions of N-ras, c-myc, c-abl and c-sis were high in two patients with CML megakaryoblastic crisis, and in one of them the c-fms was over-ex-pressed as well. The c-myc and c-fms were over-expressed in one patient with RAEB trans-formed ANLL-M7. For two patients with ANLL-M7, c-myc and c-fms mRNA were detected as well. Celi cycle and ploidy analysis by FACS showed that S phase celis were increased in four patients, and immature megakaryoblastic celis (>8N) were detected in one patient. This study emphasized that these oncogenes might be involved in the pathogenesis of megakaryocytopoietic malignant clonal proliferation.
