固相放射免疫试验检测鼠疫F1抗体

摘    要:建立了固相放射免疫试验(SPRIA)检测鼠疫F1抗体的方法。被测抗体经F1抗原两次选择结合,提高了检测特异性。检测人和5种非鼠疫疫区动物血清205份,以及假结核菌等6株非鼠疫菌免疫动物血清,全部阴性。检测鼠疫康复人和接种鼠疫苗的7种动物血清8份,IHA和SPRIA全部阳性,放射免疫沉淀(SPA—RIP)有两种动物为阴性结果;阳性反应GMT、SPRIA比IHA高,比SPA—RIP低。试验表明SPRIA检测鼠疫F1抗体,具有良好的特异性和敏感性,操作简便、快速,可检测各种标本。

Abstract:Solid-phase radioimmunoassay (SPRIA) for detecting Fl antibody of Y. pestis was established and specificity was increased by coagulating the antibody with Fl antigen twice. Results were negative when 205 sera of human and 5 kinds of animals from plague-free areas were tested and, results were also negative when sera of animals inoculated with 6 strains of non-y. pestis, including Y. pseudotuberculosis were examined. One serum of human recovered from plague and 7 sera of animals vaccinated with Y. pestis were tested by IHA, SPRIA and SPA-RIP, positive results were obtained by the former 2 methods while 2 sera were negative bv the latter. In all the tests. GMT value of SPRIA was greater than that of IHA but smaller than that of SPA-RIP. It was suggested that SPRIA was a simple, rapid method with high specificity and sensitivity in Fl antibody detection.
