热休克蛋白70在大鼠牙髓炎中的免疫组化研究

目的　研究内毒素 (LPS)诱导大鼠牙髓炎模型中诱导型热休克蛋白 (HSP) 70在不同时间动态表达及定位情况 ;推测其在牙髓炎发生、发展中的意义。方法　通过对LPS诱导的大鼠磨牙实验性牙髓炎组织进行连续切片 ,同时行超敏免疫组化分析。结果　 1d组、3d组HSP70在成纤维细胞、血管壁及内皮细胞中均呈强阳性表达 ,成牙本质细胞呈中度阳性表达 ,随后表达逐渐减弱 ,至 2周修复期为弱阳性表达。 2周以后 ,成牙本质细胞 ,前期牙本质细胞层仍有阳性表达。 3周、4周以后 ,成牙本质细胞、牙髓成纤维细胞、修复性牙本质呈弱阳性表达。 5周以后 ,牙髓组织变性、坏死 ,呈均质弱阳性染色。结论　HSP70在牙髓炎早期能够保护牙髓细胞 ,限制炎症发展 ,在损伤晚期则可能参与牙齿矿化修复过程。

An Experimental Study on the Dynamic Expression of HSP70 in Rat Pulpitis

Objective To study the dynamic expression and location of HSP27 and HSP70 in rat pulpitis model at different time and evaluate the role of HSP70 in pulp injury and reparation in rat endotoxic pulpitis. Methods Wistar rats' endotoxic pupitis model were established by application of LPS. The slides were made and examined immunohistochemically. The dynamic expression and location of HSP70 were observed. HSP70 were detected with a mouse HSP70 monoclonal antibody. Results HSP70 was strongly positive in the pulp fibroblasts,endothelial and smooth muscle cells of vessel walls. Odontoblast was moderately postive and became less gradual,and was weakly positive in 2 weeks.After 2 weeks,it was still positive in odontoblast,odontoblastic process,fibroblast and reparative dentin.Up to 3 weeks and 4 weeks,it was weakly positive in odontoblast,fibroblast and reparative dentin. Conclusion HSP70 could protect pulp cells and inhibit the development of inflammation in the early stage. HSP70 might take part in reparative dentin forming in the later stage.
