摘 要: | 目的 评价窒息性心跳骤停大鼠复苏后肾组织水通道蛋白3(AQP-3)表达的变化.方法 成年雄性Wistar大鼠20只,体重220~280 g,随机分为对照组(C组)和复苏组(R组),每组10只:R组建立窒息性心跳骤停模型,复苏采用心脏按压160次/min,静脉注射碳酸氢钠0.1 mEq/100 g+肾上腺素5μg/100 g+生理盐水0.5 ml/100 g.C组行机械通气但不建立心跳骤停模型.R组于复苏成功后1 h(T1)、3 h(T2)随机处死5只大鼠,取下腔静脉血测定血清肌酐、尿素浓度;取右侧肾脏,Westernblot法检测肾组织AQP-3表达.结果 R组T1、T2时复苏时间、血清肌酐、尿素差异无统计学意义(P0.05);与C组比较,R组T1、T2时AQP-3表达下调(P<0.05);与T1时比较,R组T2时AQP-3表达下调(P<0.05).结论 窒息性心跳骤停大鼠复苏后肾组织AQP-3表达下调早于血液肌酐、尿素浓度的改变,提示肾组织AQP-3表达反映肾功能改变的灵敏度较高.

The change in renal aquaporin-3 expression after cardiopulmonary resuscitation in rats |

Abstract: | Objective To evaluate the change in renal aquaporin-3 (AQP-3) after carcliopulmonary resuscitation ( CPR). Methods Twenty adult male Wistar rots weighing 220-280 g were randomly allocated into 2 groups ( n = 10 each) : control group (C) and CPR group. All animals were anesthetized with intraperitoneal 10% chloral hydrate 300 mg/kg, tracheostomized and mechanically ventilated. PaCO2 was maintained at 35-45 nun Hg. Femoral artery and vein were cannulated for BP monitoring and drug administration. The animals were paralyzed with vecoronium 0.1 mg/kg. Cardiac arrest was induced by suffocation and confirmed by ECG or MAP < 10 mm Hg. Cardiac arrest was maintained for 8 min before CPR was started by closed chest cardiac massage. Recovery of spontaneous circulation was defined as spontaneous rhythmic heart contraction and MAP 60 mm Hg. Five animals were killed at 1 h (T1) and 3 h (T2) after successful resuscitation. Serum creatinine and urea nitrogen concentrations and renal AQP-3 expression were determined. Results There were no significant differences between T1 and m2 serum creatinine and urea nitrogen concentrations in CPR group. The renal AQP-3 expression at T1 was significantly lower in CPR group than in control group and was significantly lower at T2 than at T1 in CPR group. Conclusion Cardiac arrest and CPR can lead to down-regulation of renal AQP-3 expression. |
