来曲唑法建立PCOS大鼠模型的评价

摘    要:目的:采用来曲唑灌胃法建立PCOS大鼠模型,并对模型进行全面的评价。方法:选取60只6周龄清洁级SD雌性大鼠,随机分为实验组和对照组各30只,采用来曲唑灌胃诱导PCOS大鼠模型;观察大鼠体重增长、动情周期变化、卵巢形态学和组织学改变;放射免疫法测定大鼠血清性激素水平;电阻抗法测定大鼠血常规;终点法测定大鼠血清生化指标。结果:①动情周期:实验组大鼠灌胃12日后失去了规律的动情周期,实验组大鼠体重增长量明显高于对照组(P<0.05)。②卵巢体积和相对质量:实验组大鼠卵巢的体积和相对质量均明显大于对照组(P<0.05)。③卵巢形态学和组织学:实验组大鼠卵巢表面苍白,体积明显增大,可见大量典型囊状卵泡;组织学检查颗粒细胞层明显减少,白膜增厚,间质增生。④血清激素水平:实验组大鼠黄体生成素(LH)、睾酮(T)浓度明显高于对照组,实验组大鼠卵泡刺激素(FSH)、雌二醇(E2)、孕酮(P)水平明显低于对照组,差异均有统计学意义(P<0.05)。⑤血常规指标:实验组大鼠血红蛋白浓度和红细胞数量均明显大于对照组(P<0.05),白细胞和血小板数量差异无统计学意义(P>0.05)。⑥血清生化指标:实验组大鼠空腹血糖(FPG)、总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)浓度明显大于对照组(P<0.05),两组大鼠血清丙氨酸氨基转氨酶活性(ALT)、天门冬氨酸氨基转氨酶活性(AST)、高密度脂蛋白胆固醇(HDL-C)、甘油三酯(TG)浓度的差异无统计学意义(P>0.05)。结论:来曲唑灌胃法可诱导出较为成功的大鼠模型,PCOS导致大鼠内分泌和代谢指标发生紊乱。

Evaluated study on the letrozole-induced model of polysystic ovarian syndrome in rats

Abstract:Objective:To establish rat models for the study of polycystic ovarian syndrome by using letrozole,and evaluated them comprehensively. Methods:Sixty SD female rats aged six weeks were divided into two groups randomly,including experimental group of thirty rats and control group of thirty rats.During the period,rats were weighted every three days;vaginal smears were collected daily for estrus cycle determination;morphologic and histologic changes of ovaries were examined after executed;Serum hormone levels were examed by RIA;Blood routine test and biochemical indexes were measured by coulter principle and end-point assay respectively. Results:①Oestrous cycle: the experimental group lost regular estrus cycle after 12 days;the increased weight of experimental rats were higher than controls singnificantly(P<0.05).②Volume and relative quality of ovaries: the volume and relative quality of ovaries from experimental group were larger than control group obviously(P<0.05).③Morphology and histology of ovaries: the ovaries of experimental rats in letrozole group seemed paler,larger and polycystic;histology showed less granular cell layer,thickened albuginea together with proliferated mesenchyma.④The serum hormone level: luteinizing hormone(LH) and testosterone(T) in letrozole experimental group were significantly higher than controls(P<0.05),serum follicle stimulating hormone(FSH),estradiol(E2) and progesterone(P) in experimental group was significantly lower than the controls(P<0.05).⑤The index of Blood routine test: the experimental rats had a singnificantly higher contents of Hb and RBC than the controls(P<0.05),no obvious difference were found with platelet between two groups(P>0.05).⑥Biochemical indexes: fasting blood glucose(FPG),total cholesterol(TC),low density lipoprotein(LDL-C) in experimental group was significantly higher than the controls(P<0.05),there were no signifancant difference with glutamic-pyruvic transaminase(ALT),glutamic-oxalacetic transaminease(AST),high density lipoprotein(HDL-C)and triglycercide(TG)(P>0.05). Conclusion:Letrozole-induced rat model is successful,PCOS can cause endocrinology and metabolism of rats to be in chaos.
