白藜芦醇对脂多糖诱导的H9c2心肌细胞损伤的影响

摘    要:目的 探讨白藜芦醇对脂多糖诱导的H9c2心肌细胞损伤的影响及其与Sirt1表达量的关系.方法 H9c2心肌细胞株培养24 h后随机分为4组:空白对照组(CON组)、白藜芦醇对照组(RES组)、模型组(LPS组)和白藜芦醇处理组(RL组).RES组和RL组细胞接受白藜芦醇处理24 h,此后LPS组和RL组细胞接受脂多糖处理24 h.处理完成后,采用乳酸脱氢酶检测试剂盒测定细胞培养液中乳酸脱氢酶含量;硫代巴比妥酸反应法检测细胞内丙二醛(MDA)的含量;荧光探针标记法检测细胞内活性氧(ROS)的含量;Western blot法检测Sirt1表达量.结果 与CON组比较,LPS组中H9c2心肌细胞乳酸脱氢酶漏出量和细胞内MDA及ROS含量显著升高,Sirt1表达量显著降低;与LPS组比较,RL组中乳酸脱氢酶漏出量和细胞内MDA及ROS含量显著降低,Sirt1表达量显著升高.结论 白藜芦醇降低脂多糖诱导的H9c2心肌细胞氧化损伤,该作用可能与白藜芦醇提高受损心肌细胞内Sirt1的表达量有关.

Abstract:To investigate the effect of resveratrol on lipopolysaccharide-induced impairment of H9c2 cardiac myoblasts and its relevance to Sirt1 expression.Methods After cultured for 24 h,H9c2 cells were randomly divided into 4 groups:control group (CON),resveratrol-treated control group (RES),model group (LPS) and resveratrol- and lipopolysaccharide-treated group (RL).Cells in group RES and RL were treated with resveratrol for 24 h while cells in group CON and LPS were treated with vehicle at equal volume.After that,cells in group LPS and RL were treated with lipopolysaccharide for 24 h while cells in group CON and RES were treated with vehicle at equal volume.Lactate dehydrogenase in the culture was determined with Lactate Dehydrogenase Detecting Assay Kit.Malondialdehyde was detected by means of reacting with thiobarbituric acid.Reactive oxygen species in cells was detected with Fluorescence Probe Labeling Reactive Oxygen Species Assay Kit.The expression of Sirt1 was determined with Western Blot.Results Compared with in group CON,lactate dehydrogenase release,malondialdehyde and reactive oxygen species content in group LPS were substantially increased while Sirt1 expression was markedly decreased.Compared with in group LPS,lactate dehydrogenase release,malondialdehyde and reactive oxygen species content in group RL were remarkably decreased,accompanied by Sirt1 expression being significantly increased.Conclusion Resveratrol attenuates lipopolysaccharide-induced oxidative injury in H9c2 cardiac myoblasts,which is probably mediated by resveratrol-induced improvement of Sirt1 expression in lipopolysaccharide-stimulated cells.
