协同凝集试验检测轮状病毒抗原的研究

作者收集346份粪便标本,进行协同凝集试验(CoA)检测轮状病毒(RV)抗原的研究,结果表明建立的 RV-CoA 有较好的特异性、敏感性和重复性,粪便标本用氯仿抽提、10%SPA 稳定液吸收、再与1%白蛋白0.1%吐温20混匀后进行 RV-CoA 检测,能有效控制非特异性凝集。RV-CoA 的阳性率与电镜法和 PAGE 法相比,无显著差异,但明显高于 ELISA 法。在 RV 流行季节,应用 RV-CoA 进行婴幼儿 RV 腹泻的快速诊断,可在40min 内出结果,阳性率为68.91%。RV-CoA 简便、快速、经济,适用于基层医院开展实验诊断和现场流行病学调查。

EVALUATION OF CO-AGGLUTINATION TEST FOR DETECTION OF ROTAVIRUS ANTIGEN

A co-agglutination test for detection of rotavirus antigen in human faecal specim- ens was developed and evaluated.A total of 346 faecal speeimens were examined.The non-specific agglutination of the test was under efficient control after the specimens were extracted with chloroform,absorbed with 10% suspension of SPA-containing sta- phylococci,and mixed with 1% bovine serum albumin 0.1% Tween 20.The positive rate of the co-agglutination test was not significantly different from that of electron microscopy or polyaerylamide gel electrophoresis,but much higher than that of enzyme- linked immunosorbent assay.In the epidemic season of rotavirus infection,the co-agg- lutination test was applied to rapid diagnosis of rotavirus diarrhoea of children (usually less than 40 minutes ).Of the 202 specimens collected in the epidemic season in 1989,123 were positive (68.91%).The results obtained in this study indicate that the co-agglutination test has a specificity,sensitivity and reproducibility in addition to its Simplicity,rapidity,convenience and economy.It may be applied for small hospitals to make a laboratory diagnosis or epidemiological investigation on the spot.
