摘    要:目的:观察激素(地塞米松)预处置对兔油酸型急性呼吸窘迫综合症【ARDs)血清TNF—a、IL-8和IL-10的影响,探讨激素治疗AP.DS的理论基础。方法:经兔耳缘静脉注射油酸复制兔ARDS模型,用酶联免疫吸附试验测定血清中TNF—a、IL-8和IL-10水平,结合血气分析检测及肺组织病理学改变进行观察分析。结果:ARDS组兔血清TNF—a、IL-8水平明显高于对照组(P〈0.01);IL-10水平明显低于对照组(P〈0.01)。地塞米松预处置组血清IL-10水平有上升恢复,高于ARDS组(P〈0.01)。地塞米松预处置组血清TNF—a、IL-8水平降低,低于ARDS组(P〈0.01)。动脉血气分析及肺组织病理学有相应改变。结论:TNF-a、IL-8和IL-10在兔油酸型ARDS炎症中可能起重要作用,地塞米松可能通过抑制TNF-a、IL-8过度分泌,促进IL-10合成而减轻肺损伤。

Abstract:Objective: To observe the effects mechanism of glueocorticoids (dexamethasone) pretreatment on II, -6, IL-8, TNF-a, IL-IO levels in serum of rabbits with oleic acid ARDS. Methods: rabbits' ARDS model was repli- cated through injecting oleic acid to rabbits' veins under ears and the levels of IL-8, TNF-a, IL-10 in serum was measured using enzyme linked immunoserbent assay (ELISA). Results: In ARDS group the levels of IL-8, TNF- a in serum were significantly higher than that of the control group(P〈O. O1 ). wheras the levels of IL-IO in serum was no- ticeably lower than that of the control group (P(O. 01) .In dexamethasone pretreatment group the levels of IL-8, TNF-g in serum were significantly lower than that of the ARDS group (P 〈 0. 01 ) whereas the levels of IL- 10 in serum was significantly higher than that of the ARDS group (P〈0. 01 ). Conclusions: IL-8, TNF-a, IL-IO may play important roles in the inflammatory process of oleic acid ARDS in rabbits while the dexamethasone may relieve lung injuries by inhibiting excessive discharging of IL-8 and TNF-a.
