长期服用辛伐他汀对机械通气肺损伤大鼠肺湿/干重比与蛋白含量的影响

摘    要:目的对长期服用辛伐他汀大鼠进行机械通气造成急性肺损伤,观察大鼠肺组织病理学变化、湿/干重比(W/D)及蛋白含量的变化,探讨长期服用辛伐他汀对机械通气致急性肺损伤、肺水肿及肺泡通透性的影响。方法32只雄性SD大鼠随机分为4组(n=8):对照组(A组)不做任何处理;机械通气组(B组)行机械通气,潮气量7ml/kg;辛伐他汀组(C组)进行辛伐他汀(10mg·kg~·d^-1)灌胃处理4周;辛伐他汀+机械通气组(D组)辛伐他汀灌胃4周后行机械通气,潮气量7ml/kg。B组与D组行机械通气4h后处死大鼠,收集肺组织,苏木精一伊红染色光镜下观察病理学改变,测定肺组织W/D比及蛋白含量。结果与A组相比,B组大鼠肺组织损伤明显,W/D与蛋白含量均明显增高(P〈0.01);C组各指标无明显变化。与B组相比,D组肺组织病理损伤明显减轻,W/D及蛋白含量明显降低(P〈0.01)。结论长期服用辛伐他汀可以明显降低肺血管与肺泡的通透性,减轻机械通气所致的急性肺损伤、肺水肿。

Abstract:Objective To investigate the impact of long - term use of simvastatin on wet - to - dry weight ratio ( W/ D) and protein content in ventilator - induced lung injury in rats. Methods Thirty - two male SD rats were randomly divided into four groups (n = 8 each) : group A received room air without mechanical ventilation; group B received tidal volume of 7 ml/kg with mechanical ventilation for 4 h; group C : animals were orally administered with 10 rag/( kg . d) simvastatin for 4 weeks; group D: animals were orally administered with 10 mg/(kg . d) simvastatin for 4 weeks and then received tidal volume of 7 ml/kg with mechanical ventilation for 4 h. After 4 h of ventilation, the rats were sacrificed and the lung tissue were collected and stored with correct methods. Pathological changes in the lung was observed by mi- croscope. The measured indexes included W/D of lung and the levels of protein content in lung tissue. Results Pathological damage of lung tissue in group B was significanly more severe than in group A. The levels of protein content in lung tissue and W/D in group B were significantly higher than in group A ( P 〈 0.01 ). Compared with group B, those indexes in group D were significantly decreased ( P 〈 0.01 ). Conclusion Long - term use of simvastatin can significantly reduce pulmonary vascular and alveolar permeability and lighten pulmonary edema c causedventilation in acute lung injury.
