依达拉奉对脑缺血/再灌注损伤大鼠Caspase-3表达的影响

摘    要:目的探讨依达拉奉对急性脑缺血/再灌注(I/R)损伤大鼠的保护作用。方法制作线栓法大鼠局灶性脑缺血/再灌注模型,72只SD大鼠随机分为假手术组(Sham组)、脑缺血/再灌注组(Mod组)、依达拉奉组(Eda组),每组再分为再灌注2h,4h、12h、24h4个亚组,每个亚组6只大鼠。各时间点进行大鼠神经功能缺损评分(NDS),测定血清Bax数值,计数海马CAl区的神经元Caspase-3蛋白表达阳性细胞数,观察12h组组织病理学改变。结果(1)Sham组大鼠无神经功能缺损,NDS为0;Mod组和Eda组大鼠均有明显的神经功能缺损,与Mod组比较,Eda组NDS显著降低(P〈0.05)。②各时间点Mod组和Eda组Bax和Caspase-3的表达均显著高于Sham组(P〈0.05);与Mod组比较,Eda组Bax和Caspase-3的表达显著降低(P〈0.05)。结论依达拉奉能减轻局灶性脑缺血/再灌注大鼠的神经功能缺损,大鼠肢体运动功能得到改善。其对脑缺血/再灌注损伤的保护作用可能通过抑制Bax和Caspase-3的表达来实现。

Effect of edaravone on the expression of Caspase-3 in rats with cerebral ischemia/reperfusion injury

Abstract:Objective To investigate the protective effect of edaravone in rats with acute cerebral ischemia/reperfusion injury. Methods The middle cerebral artery occlusion (MCAO) model was established on 72 SD rats, which were randomly divided into 3 groups: sham operation group (Sham group), ischemia/reperfusion group (Mod gonp) and pre -edaravone group (Eda group). According to the repeffusion time, each group was randomly divided into 2 h, 4 h, 12 h, and 24 h subgroup,6 rats in each subgroup. After ischemia/reperfusion, the neurologic deficit score (NDS) was evaluated. The Bax concentration of blood was determined and the expression of Caspase - 3 by the positive - stained ceils in hippocampal CA1, and histopathological changes in each group at 12 h were observed. Results (1) The NDS in Sham group was 0, while the NDS in Mod group and Eda group was higher than that of Sham group. Compared with Mod group, the NDS was significantly lower in Eda group (P 〈 0.05 ). (2) Compared with Sham group, the concentration of Bax and the number of positive expression of Caspase - 3 were significantly higher in Mod group and Eda group ( P 〈 0.05 ) at all time points. The expression of Bax and Caspase - 3 was significantly down - regulated in Eda group than in Mod group ( P 〈 0. 05). Conclusions (3) Edaravone can reduce NDS in rats with focal cerebral ischemia/reperfusion, and can improve limb movement function ; @ Edaravone can inhibit the expression of Bax and Caspase - 3 resulted from ischemia/ reperfusion, which may suggest that both of them play an important role in acute cerebral ischemia/reperfusion injury. Edaravone can protect neurons from ischemic damage by down - regulating the expression of Bax and caspase - 3.
