大鼠松果体星形胶质细胞与少突胶质细胞的增龄变化

摘    要:目的:探讨增龄变化对大鼠松果体内星形胶质细胞和少突胶质细胞的影响。方法:20只SD大鼠,随机分为青年组(4月龄组)和老年组(24月龄组),每组10只。采用Benda氏胶质细胞染色,分别进行胶质纤维酸性蛋白(GFAP)与半乳糖脑苷脂(GC)免疫组化染色,显微镜下观察松果体内星形胶质细胞和少突胶质细胞并计量分析。结果:随年龄增长松果体胶质细胞主要变化:1.松果体内星形胶质细胞体积增大,GFAP阳性细胞数增多(P〈0.05),阳性产物平均光密度值增高(P〈0.05);2.GC阳性细胞数减少,阳性产物平均光密度值降低(P〈0.05)。结论:大鼠松果体随增龄星形胶质细胞增多、少突胶质细胞减少,其变化可能是导致机体发生衰老的原因。

Abstract:Objective: To observe the changes on astrocyte and oligodendeoeyte in the pineal body with aging in rats. Methods: 20 Sprague -Dawley rats were used in the experiment and randomly divided into two groups with 10 in each group: the youth group (4 months) and the old - age group (24 months) . Benda neuroghal cell staining, Glial Fibrillary Acidie Protein (GFAP) and Gallc to Cerebroside (GC) immunohis- tochemistry staining were used for light microscopic observation. The quantity of on glial cell, GFAP - positive cells and CA: - positive ceils in the pineal body of rats were quantitatively analyzed with cell morphometrie technique. Results: 1. the quantity and volume of neuroglial cell were increased, simultaneously the numbers and average optical density of GFAP - positive cells in the pineal body were significantly increased ( P 〈 0. 05) ; 2. the numbers and average optical density of GC - positive cells were significantly decreased in the pineal body with aging ( P 〈 0. 05 ) . Conclusion: The quantities of astrocyte were significantly increased, but the oligodendeocytes were decreased in the pineal body with aging. It might be one of the mechanisms on structural changes of pineal body would be one cause of the body taked place the decrepitude.
