摘 要: | 目的观察青蒿提取物对房颤大鼠心肌炎症相关因子肿瘤坏死因子-α(TNF-α)及细胞间黏附分子-1(ICAM-1)表达的影响,探讨青蒿抗心律失常的作用机制。方法将大鼠分为对照组、模型组、青蒿组、卡托普利组。采用氯化钙、乙酰胆碱混合液以舌下静脉给药的方式建立房颤动物模型,隔日重复1次,连续4周,停药后心电图仍有典型房颤改变为造模成功。实验当日,动物静脉血,ELISA法测样品中TNF-α、ICAM-1浓度。结果模型组血清TNF-α、ICAM-1水平较对照组均有提高(P<0.05)。卡托普利组在降低TNF-α、ICAM-1表达方面作用最为显著,青蒿组作用略差于卡托普利组(P>0.05)。结论青蒿提取物通过抑制炎症因子TNF-α、ICAM-1,抑制心脏结构重构和电重构,减少心律失常的发生。

Abstract: | Objective To observe the effect of artemisia annua L.extract(AALE) on expression of tumor necrosis factor-α(TNF-α) and intercellular adhesion molecule-1(ICAM-1) in rats with atrial fibrillation,and to explore anti-arrhythmic mechanism of artemisia annua.Methods Sixty Sprague-Dawley(SD) rats are divided into the control group(n=15),the model group(n=15),AALE group(n=15),and captopril group(n=15).Models with atrial fibrillation were established by sublingual intravenous administration of calcium chloride and acetylcholine mixture.The concentrations of serum TNF-α and ICAM-1 were tested by ELISA.Results The levels of serum TNF-α and ICAM-1 in model group were significantly increased than that in the control group(P<0.05).The expression of TNF-α and ICAM-1 in captopril group was most significant,and that in AALE group was slightly lower than that in captopril group.There was no significant difference between these two groups(P>0.05).Conclusion Artemisia annua L.extract might inhibit cardiac structural remodeling and electrical remodeling,and reduce the incidence of arrhythmias by inhibiting the inflammatory cytokines TNF-α,ICAM-1. |
