肢体缺血预处理对大鼠全肝缺血再灌注肠黏膜屏障功能的影响

摘    要:【目的】观察肢体缺血预处理对SD大鼠全肝缺血再灌注肠黏膜屏障功能的影响。【方法】24只SD大鼠随机分成假预处理组(S组,n=8)、全肝缺血再灌注组(IR组,n=8)、肢体缺血预处理组(LIP组,n=8)。S组行假预处理及24h后行假手术;IR组行假预处理及24h后行全肝缺血再灌注;LIP组分离双下肢股动静脉后阻断5min再开放10min,重复3次,24h后行全肝缺血再灌注。3组动物于再灌注3h或者相应时间点取回肠末端组织和肠系膜上静脉血,检测回肠末端二胺氧化酶(DAO)活性和血浆D-乳酸、内毒素含量。【结果】与S组相比,IR组和LIP组回肠末端DAO活性显著降低(P〈0.05),血浆D-乳酸、内毒素含量显著升高(P〈0.05);LIP组回肠末端DA0活性显著高于IR组,血浆D-乳酸、内毒素含量低于IR组(P〈0.05)。【结论】肢体缺血预处理能够保护SD大鼠全肝缺血再灌注时损伤的肠黏膜屏障功能。

Effect of Limb Ischemic Preconditioning on Intestinal Mucosal Barrier Function Following Total Hepatic Ischemia-reperfusion in Rats

Abstract:[Objective]To observe the effect of limb ischemic preconditioning on intestinal mucosal barrier function following total hepatic ischemia-reperfusion in rats. [Methods]Twenty four SD rats were divided randomly into the sham preconditioning group(Group S, n = 8), total hepatic ischemia-reperfusion treatment group(Group IR, n = 8) and limb ischemic preconditioning group(Group LMP, n = 8). Group S received sham limb ischemic preconditioning and sham operation 24h later. Group IR received sham limb ischemic preconditioning and total hepatic ischemia-reperfusion 24h later. Group LIP were performed with separated bilateral hind limb femoral arteries and veins for three alternate cycles of 5min ischemia followed by 10min reperfusion, then followed by total hepatic ischemia-reperfusion for 24h. Terminal ileum tissue and blood in the superior mesenteric vein were collected at 3h after reperfusion or corresponding time point. Activities of terminal ileum tissue diamine oxidase(DAO), and the plasma concentration of D-lactate and endotoxin were measured. [Results]Compared with Group S, the activities of terminal ileum tissue DAO in Group IR and Group LIP were significantly lower( P〈0.05), and the plasma concentration Of D-lactate and endotoxin were significantly higher( P 〈0.05). The activities of terminal ileum tissue DAO in Group LIP were significantly higher than those of Group IR( P 〈0.05), and the plasma concentration of D-lactate and endotoxin were significantly lower. [Conclusion]Limb ischemic preconditioning has protective effect on intestinal mucosal barrier function fol- lowing total hepatic ischemia-reperfusion in SD rats.
