摘    要:目的本实验拟采用全细胞膜片钳记录技术,研究马桑内酯(CL)对培养大鼠皮层神经元瞬时外向钾电流(ⅠA)的影响,从而探讨CL致癫痫的电生理机制及与ⅠA的关系,为临床更有效地防治癫痫提供新的依据。方法采用全细胞膜片钳技术记录ⅠA,进行以下方面实验①培养大鼠皮层神经元ⅠA的特性及鉴定;②三种不同浓度CL对神经元ⅠA的影响;③CL对神经元ⅠA的I-V曲线的影响。结果测试电压为+65mV时,三种不同浓度CL后均可抑制IA的电流振幅,其抑制差异均有统计学意义。结论CL具有抑制瞬时外向钾电流的作用,且在一定范围内抑制呈剂量依赖性,推测CL致癫痫的发生机制可能与IA的抑制有关。

Abstract:Objective The objective is to observe the effect of coriaria lactone (CL) on the cortex neuron I_A of cultured newborn rat,so as to find out the electrophysiological mechanism of CL-induced epilepsy and offer a new experimental basis for preventing and treating epilepsy.Methods I_A was elicited by application of voltage-clamp steps from holding potential of-65 mV to 95 mV.I_A was respectively tested after using CL in three concentrations of 5,50,100 mg/L.Results The patch clamp experimental results demonstrated that I_A was decreased by 5,50 or 100 mg/L CL.Conclusion The results indicate that CL has an action of decreasing I_A;and the effect may be related to CL-induced epilepsy.
