摘    要:研究目的探讨蝙蝠葛苏林碱(DSL)对局部脑缺血-再灌注损伤的治疗作用。研究背景已确证某些钙通道阻断剂(如尼莫地平)有抗脑缺血作用。DSL是新近证明的、能选择性作用于神经细胞的钙通道阻断剂,它对脑缺血的影响尚未见研究报道。研究方法 40只wistar雄性大鼠随机分为5组。Ⅰ、Ⅱ、Ⅲ是低、中、高3个不同剂量DSL的实验组;Ⅳ为阴性对照组;Ⅴ为尼莫地干阳性对照组。用尼龙线经颈内动脉可逆性插入到大脑前动脉内,建立大鼠局部脑缺血再灌注模型。脑电图在双侧顶区记录。研究结果 MCA阻断后,缺血侧的EEG波幅降低,甚至成电平线,然后逐渐恢复,1小时后可恢复到50%以上。复通后1小时Ⅱ、Ⅲ及Ⅴ组的EEG波幅有明显增高,与阴性对照的Ⅳ组差异显著(P<0.05),且这3组动物的生存时间也明显延长。结论 DSL对局部脑缺血-再灌注损伤有一定的保护作用。

Abstract:Objective To search the therapeutic effectsof daurisoline for injury of focal cerebralischemia—reperfusion. Background It has been proved thatCa”channel beockers such as nimodipine can im.prove the conditions of cerebral ischemia.Danrisoline which was synthesised lately was aCa斗channel blocker acted selectively on neuron.But its effects on cerebral ischemia has not beenstudied. Methods Forties wistar rats was divided jn.to five groups randomly.Group l,group 2 andgroup 3 experimental groups which treated withdaurisoline.Group 4 was negative contr01.Group5 was positive control treated with nimodipine.Focal cerebral ischemia—’reperfusion was devel—oped in rats by reverbly inserting a nylon threadinto the anterior cerebral artery through the inter·nal carotid artery. Electroencephalogram(EEG)was recorded on both sides of skull. Results After middle cerebral arteryocclusion,EEG became lower,SO far as to elec—trical plane line,then restored progressively andreached above 50%normal in l hour.One hourafter reperfusion,the range of EEG in group 2,group3,.and group 5 increased more than that ingroup 4(P<0.05),and the life—span in group2,group 3,and group 5 was longer than that ingroup4. Conclusions Daurisoline may protect brainagainst injury of ischemia and reperfusion to someextent.
