防己黄芪汤对阿霉素肾病大鼠蛋白尿及炎性因子的影响

摘    要:目的:考察防己黄芪汤改善阿霉素肾病大鼠蛋白尿的效应强度以及对炎性因子的影响,探讨防己黄芪汤调节蛋白尿的作用机制.方法:按5mg/kg体重给予大鼠尾静脉1次注射阿霉素建立阿霉素肾病模型;生化分析仪检测24h尿蛋白定量,ELISA法检测肾组织IL-18、TGF-β1、CTGF的浓度.结果:模型组大鼠24h尿蛋白定量及肾组织IL-18、TGF-β1、CTGF浓度均升高(P<0.05);防己黄芪汤高、中、低剂量组尿蛋白定量明显减少(P<0.05),肾组织IL-18、TGF-β1、CTGF浓度降低(P<0.05).结论:防己黄芪汤改善阿霉素肾病大鼠蛋白尿的作用,可能与其降低肾组织IL-18、TGF-β1、CTGF浓度有关.

Abstract:Objective:To investigate the mechanism of Tetrandra and Astragalus decoction for regulating proteinuria,through studying its effect on improving proteinuria and impacts on inflammatory factors in nephrotic rats induced by adriamycin.Methods:The tail vein of the rat was injected with adriamycin by 5mg/kg to establish an adriamycin nephropathy model.24-hour urinary protein excretion was tested by using the biochemical analyzer,and the concentrations of kidney tissue IL-18,TGF-β1 and CTGF were assayed by using the ELISA kits.Results:In model rats,24-hour urinary protein excretion and the concentrations of kidney tissue IL-18,TGF-β1 and CTGF were higher (P < 0.05) ;in Tetrandra and Astragalus decoction groups with high-dose,medium-dose and low-dose,urinary protein excretion decreased significantly (P < 0.05),and the concentrations of kidney tissue IL-18,TGF-β1 and CTGF were lower (P <0.05).Conclusion:Tetrandra and Astragalus decoction for improving proteinuria in rats with adriamycin nephropathy may be related to reducing the concentrations of kidney tissue IL-18,TGF-β1 and CTGF.
