苓桂术甘汤预防性给药对心肌缺血再灌注损伤NO、NOS含量的影响

摘    要:目的:通过苓桂术甘汤预防性给药,观察心肌缺血再灌注损伤家兔血清一氧化氮(NO)和一氧化氮合酶(NOS)含量的变化,探讨其防治心肌缺血再灌注损伤的可能机制。方法:40只健康家兔随机分成5组,结扎左冠状动脉前降支制备心肌缺血再灌注损伤模型,再灌注3h后采血并检测血清中NO和NOS含量,取左心室心肌组织做病理切片。结果:各组NO及NOS含量比较,假手术组与空白组间无统计学差异(P>0.05);模型组NO及NOS含量明显下降,较假手术组具有统计学差异(P<0.01);高剂量组、低剂量组分别与模型组比较,NO及NOS含量均显著升高,具有统计学差异(P<0.01),且病理结果显示,高剂量组疗效优于低剂量组.结论:苓桂术甘汤预防性给药对心肌缺血再灌注损伤具有保护作用,其可能机制与升高NO和NOS含量有关,且高剂量组疗效显著优于低剂量组。

Effect of Prophylactic Administration of Linggui Zhugan Decoction on NO,NOS Caused by Myocardial Ischemia -reperfusion Injury

Abstract:Objective : Through prophylactic administration of Linggui Zhugan decoction for myocardial ischemia- reperfusion injury, to observe content changes of nitric oxide (NO) and nitric oxide synthase (NOS) in rab- bits serum, and to explore its possible mechanism preventing myocardial ischemia -reperfusion injury. Meth- ods: 40 healthy rabbits were randomly divided into five groups, ligating the left anterior descending coronary artery for the preparation of myocardial ischemia - reperfusion injury models, and examine the contents of NO and NOS in the serum, and take the left ventricular myocardium tissue biopsy done after reperfusion three hours. Results : Compared with the blank group, the contents of NO and NOS in the sham group were withoutstatistically significant the model group were difference ( P 〉 0.05 ) ; compared with the sham group, the contents of NO and NOS in significantly decreased( P 〈0.01 ) ; high- doses group and low- dose group were corn- pared with the model group, and the contents of NO and NOS were significantly increased, with a statistically significant difference (P 〈 0.01 ) respectively, and cardiac pathology results showed that high - dose group was better than the low - dose group. Conclusion : Prophylactic administration of Linggui Zhugan decoction has a protective effect on myocardial ischemia - reperfusion injury models. The mechanism may be related to the increase of NO and NOS, and high - dose group is more effective than low - dose group.
