鹿茸多肽诱导心肌干细胞分化作用及对心肌细胞特征性MHC基因表达的影响

摘    要:目的从新生大鼠心脏中分离得到心肌干细胞,体外培养,逆转录聚合酶链反应(RT-PCR)法观察鹿茸多肽诱导前后心肌细胞特征性基因α-心肌肌球蛋白重链(α-MHC)和β心肌肌球蛋白重链(β-MHC)的表达,探讨鹿茸多肽对心肌干细胞分化的促进作用。方法胰酶消化法分离心肌干细胞(CSCs),并进行传代培养,流式细胞法及免疫组化染色鉴定,于第3代CSCs培养液中加入不同浓度的(50,100,200,400,800,1 600μg/mL)鹿茸多肽进行体外诱导,3周后,用细胞刷收集细胞,利用RT-PCR技术观察心肌细胞特征性基因MHC的表达情况。结果鹿茸多肽对CSCs的MHC基因表达有促进作用,在一定范围内存在量效关系,并随着鹿茸多肽浓度的增加,α-MHC和β-MHC基因的表达均有所增加。结论鹿茸多肽对心肌干细胞分化心肌细胞有促进作用,表明其在心肌损伤性疾病的康复中有潜在的治疗价值,值得进一步全面深入的研究。

Abstract:Objective To investigate the effects of velvet antler polypeptides on myocardial stem cell differentiation in vitro, and to explore the underlying mechanism. Methods The different concentration of elvet antler polypeptides(50,100,200, 400,800, 1 600 μg/mL) were injected into the third generation of myocardial cells to investigate myosin heavy chain gene expression in cardiac myoeytes. Results Velvet antler polypeptides induce MHC gene expression. With the increase of Velvet antler polypeptides, the α-MHC and β-MHC gene expression level increased. Conclusion Velvet antler polypeptides promote the gene expression. With potential therapeutic value in myocardial injury disease rehabilitation, it is worthy of further study.
