情感障碍高发家系遗传学初步研究

目的　探测儿茶酚氧位甲基转移酶 (COMT)基因、载脂蛋白E(Apo E)基因及五羟色胺受体 2A(5HT 2a)基因多态性与情感障碍之关系 ,搜寻中国汉族人情感障碍易患性基因。方法　应用聚合酶链反应 (PCR)和限制性片段长度多态性 (RFLP)方法对 5个情感障碍高发家系进行初步研究。结果　用改进的传递 /不平衡 (TDT)检测结果表明 ,在 5个情感障碍高发家系中 ,COMT基因与情感障碍相关联 (P <0 0 5 ) ,而Apo E基因和 5HT 2a基因不与情感障碍关联 (P >0 0 5 )。结论　在我们研究的家系中 ,2 2号染色体和长臂 (2 2q11 2 )可能存在情感障碍易患性基因

Preliminary genetics study of five pedigrees with high incidence of affective disorder

Objective To Explore the relationships among polymorphism of COMT gene, Apo E gene and 5HT 2a gene with affective disorder and find a gene predisposing in affective disorder in the Han nationality of Chinese. Methods Five pedigrees with high incidence of affective disorder were preliminary studied by polymerize chain reaction(PCR) and restriction fragment length polymorphism(RFLP) techniques. Results Analysis results of statistics of the transmission/disequilibrium test(TDT) handing showed that the COMT gene was associaed with affective disorder( P <0 05), the Apo E gene and the 5HT 2a gene were not associatied with affective disorder( P >0 05) in the five pedigrees with high incidence affective disorder. Conclusions Our study results suggested that there was an affective disorder liability gene on 22chromosome(22q11 2) in our studied pedigrees.
