缺氧复氧性心肌细胞损伤过程中钙敏感受体作用探讨

摘    要:目的观察钙敏感受体(CaSR)在缺氧复氧性大鼠心肌细胞损伤过程中的作用。方法将45份原代培养4d的大鼠心肌细胞按随机数字法分为正常对照组、缺氧/复氧组、钙阻断剂组、CaSR激动剂组和钙耗竭组,各9份。用饱和氮气pH6.8的D—Hands液培养心肌细胞3h,再用含20%新生牛血清的DMEM液培养心肌细胞9h,复制心肌缺氧/复氧模型。测定肌酸激酶(cK)含量。结果不同组CK含量差异有统计学意义(P〈0.01)。缺氧/复氧组、钙阻断剂组及CaSR激动剂组CK含量均高于正常组(均P〈0.05),钙耗竭组与正常组CK含量差异无统计学意义(P〉0.05);钙阻断剂组、CaSR激动剂组及钙耗竭组的CK含量均低于缺氧/复氧组(均P〈0.01);CaSR激动剂组的CK含量高于钙阻断剂组和钙耗竭组(均P〈0.05)。结论CaSR参与大鼠缺氧复氧性心肌细胞损伤。

Abstract:Objective To observe the role of calcium-sensing receptor(CaSR) in myocardial hypoxia/reoxygena- tion in rats. Methods Primary cardiomyocytes were cultured for four days, then randomly divided into five groups: control group, hypoxia/reoxygenation group(H/Re), calcium channel inhibition group, CaSR activation group and cal- cium depletion group. The cardiomyocytes were in ischemia-mimetic solution for three hours, and re-incubated in normal culture medium for nine hours to establish a model of H/Re. The levels of creatine kinase (CK) were meas- ured. Results CK level was significantly increased after hypoxia/reoxygenation. The CK level was lower in the Na+ -Ca2+ exchanger and L-calcium channel inhibition group compared with that in the H/Re group. CK level was signifi- cantly higher in the CaSR group than that in the calcium channel inhibition group and calcium depletion group (P 〈 0.05).Conclusions CaSR was involved in the hypoxiaJreoxygenation injury in rats.
