针刺对自由运行状态金黄地鼠视交叉上核内5-HT7受体功能的影响

摘    要:目的:探讨针刺导引节律的5-HT受体转导机制。方法:金黄地鼠经驯化进入自由运行状态后,于DD第三天CT9给予电针、8-OH-DPAT、光脉冲导引、捆绑导引刺激。刺激后2 h取样,应用放免法测定视交叉上核(SCN)内5-HT7受体功能的变化。结果:电针和8-OH-DPAT组KD值显著低于捆绑对照组;光脉冲导引组的KD值明显高于捆绑对照组,差异有统计学意义;说明电针和8-OH-DPAT均能使5-HT7受体的亲和力增强,而光脉冲刺激则可抑制其亲和力。结论:针刺通过提高5-HT7受体的活性,实现在主观白天的相位超前性调整。

Abstract:Objective:To research the regulating effects of electro-acupuncture on 5-HT receptors in the suprachiasmatic nucleus(SCN).Methods: After free-running,hamsters were divided into electro-acupuncture group,8-OH-DPAT group,photic stimuli group and binding group rando mLy.Electro-acupuncturing "Baihui"(Du20),"Changqiang"(Du1),system injecting 8-OH-DPAT,photic and binding stimuli were given respectively at circadian time 9(CT9) in the third day after constant darkness.The regulating effects of electro-acupuncture on 5-HT_7 receptors in SCN were detected by using radioligand binding assay of receptors.Results:The dissociation constant(KD) of electro-acupuncture and 8-OH-DPAT groups was significantly lower than that of binding group.And the KD of photic stimuli group was remarkably higher than that of binding group.It means that electro-acupuncture and system injecting 8-OH-DPAT improved affinity of 5-HT_7 receptors but photic stimuli decreased it in.Conclusion:electro-acupuncture induced phase advance at subjective day in hamster spontaneous behavior by activating 5-HT_7 receptors in SCN.
