三黄泻心汤治疗上消化道出血的药效学研究

目的：研究三黄泻心汤(SXT)对凝血系统和胃粘膜损害的影响。方法：以毛细管法和割尾法分别测定SXT对小鼠的凝血和出血时间的影响；以利血平腹腔注射(10mg/kg)造成小鼠胃粘膜损害观察SXT对抗胃粘膜损害的作用；大鼠幽门结扎收集胃液，考察SXT对胃液分泌的影响。体外实验中观察SXT对家兔血浆复钙时间、ADP诱发的血小板聚集和离体胸主动脉条收缩力的影响。结果：SXT可缩短出凝血和血浆复钙时间，促进血小板聚集，增加家兔离体胸主动脉条的收缩力，并具有对抗胃粘膜损伤和降低胃蛋白酶的作用。结论：SXT通过作用于内源性凝血系统，促进血小板聚集和血管收缩表现出明显的促凝血和止血作用，并对不良刺激所致的胃粘膜损伤具有保护作用。

Pharmacodynamic study of Sanhuang Xiexin Tang (三黄泻心汤) for treatment of upper gastrointestinal hemorrhage

Objective: Sanhuang Xiexin Tang(SXT) is a Chinese traditional medicine and widely used for the treatment of upper gastrointestinal hemorrhage and other hemorrhage disorders. We invested effects of SXT on blood coagulation and gastric mucous membrane lesion in experimental animals and tried to get a rudimentary understanding of its pharmacodynamics. Methods: We checked effects of SXT on clotting time and bleeding time in mice, plasma recalcification time, ADP induced platelet aggregation and the contractility of thoracic aorta strip in rabbits in vitro; meanwhile, we investigated protect activity of SXT in anti-gastric mucous membrane lesion induced by reserpine 10mg/kg ip in mice and its effect on gastric secretion from pylorus ligated rats. Results: SXT shortened clotting time and bleeding time in mice, accelerated platelet aggregation and increased the contractility of thoracic aorta strip in rabbits. SXT still showed the ability to protect mice from gastric mucous membrane lesion and depress pepsin activity in pylorus ligated rats. Conclution: SXT promoted the process of clotting and hemostasis, which was mediated through its effects on intrinsic coagulation system, platelet aggregation and vasoconstriction. SXT could protect gastric mucous membrane from harmful stimulations.
