海水浸泡对保存血液中红细胞质量影响的实验研究

摘    要:目的观察海水浸泡对保存血液红细胞质量的影响。探索海洋环境下血液的运输和保存。方法将全血(WB)、添加剂红细胞(AS-RBS)和去白细胞红细胞(LP-RBC)三种血液制品各20袋等量分为两份,标记为浸泡实验组与对照组,对照组采用三层软塑料膜外包装将血袋密封,空气隔绝海水;浸泡实验组不包裹直接将血袋与海水接触。两组标本同时置于同一地点、同一海水深度中浸泡72h后取出,再放置4±2℃血库冰箱保存观测35d。选取浸泡前、海水浸泡72h后及再置血库冰箱保存后每周一次分别测定各成分血液游离血红蛋白(FHb)、平均红细胞体积(MCV)、平均血红蛋白含量(MCH)、平均红细胞血红蛋白浓度(MCHC)、红细胞体积分布宽度(RDW)、红细胞及网织红细胞计数、红细胞脆性试验,同时进行细菌培养。结果海水浸泡72h后保存血液红细胞与常规4±2℃血库冰箱保存血液有相似的变化趋势,三种保存血液制品中红细胞质量变化有一定差别,其中LP-RBC变化最大。各组细菌培养均为无菌生长。结论采用相同的血液保养液袋,对血液再包装进行海水浸泡72h,入库4±2℃血库冰箱再保存对其红细胞形态和功能有一定影响,但总的变化趋势与常规4±2℃血库冰箱保存相似。

Experimental study on the changes of RBC figurations in the preserved bloods after dipping in seawater
Abstract:Objective To observe the rule of changes of RBC figurations in the preserved bloods after dipping in seawater. Methods Three kinds of blood products including WB, AS-RBC and LP-RBC were averagely divided into two groups marked experiment group and control group. Control groups wrapped with soft plastic membrane and experiment groups directly were dipped into seawater for 72h; then transferred to 4±2℃ refrigerator and continued preservation for 35days. Free hemoglobin(FHb), mean corpuscular volume(MCV),mean corpuscular hemoglobin(MCH),mean corpuscular hemoglobin concentration(MCHC),RDW and other correlated items about RBC figurations of red blood cell in all blood products were detected respectively before dipping in seawater ,three days after dipping in seawater and weekly after stored in 4±2℃ refrigerator. Results The general rule of changes of RBC figurations in preserved bloods after dipping in water 72h was almost similar as the regularly 4℃ preserved bloods, but FHb release in the LP-RBC groups were significantly more than those of WB and AS-RBC groups.Conclusion Red blood's succeeding shelf life in three kinds of blood products should be adjusted when they are transferred to 4±2℃ refrigerator after preserved in seawater 72h so that the blood quality is ensured.
