单核细胞趋化蛋白1基因多态性与2型糖尿病的相关性研究

摘    要:目的探讨中国西北地区汉族人群中2型糖尿病(T2DM)与单核细胞趋化蛋白1(MCP-1)-2518(A/G)基因多态性的关系。方法测量41例正常对照者和57例2型糖尿病患者相关临床生化指标,并采用聚合酶链反应限制性片段长度多态性(PCR-RFLP)技术,观察MCP-1-2518(A/G)基因多态性,研究其与2型糖尿病的关系。结果2型糖尿病组与正常对照组的MCP-1等位基因和基因型频率比较差异无统计学意义(P>0.05);多因素Logistic回归分析显示,MCP-1 G等位基因为2型糖尿病的保护因素(OR=0.015,CI=0.005~0.437),且有统计学意义(P<0.05)。结论正常对照组与2型糖尿病组MCP-1-2518(A/G)基因多态性差异无统计学意义,尚不能认为在中国西北汉族人群中MCP-1基因多态性与2型糖尿病之间有相关性,但G等位基因可能为2型糖尿病的保护因素,它的存在降低2型糖尿病的患病率。

Abstract:Objective To determine whether the monocyte chemoattractant protein-1(MCP-1)-2518(A/G) gene polymorphism was associated with type 2 diabetes mellitus in the north-west population of China.Methods MCP-1 polymorphism was investigated in 41 cases of normal control subjects and 57 cases of type 2 diabetes patients by technique of the polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP),and the association of MCP-1 polymorphism with type 2 diabetes mellitus was explored,plasma biological and chemical quota were measured at the same time.Results There were no significant differences in genotype frequencies and allele frequencies of MCP-1 gene between type 2 diabetes mellitus group and normal control group(P>0.05).Multivariate logistic regression analysis showed that MCP-1 G allele was a protective factor of type 2 diabetes mellitus(OR=0.015,CI=0.005-0.437,P<0.05).Conclusion MCP-1 gene polymorphism genotype frequencies and allele frequencies of type 2 diabetes mellitus patients are not significantly different from those of normal control,MCP-1 gene polymorphism is not associated with type 2 diabetes mellitus.MCP-1 G allele is a protective factor of type 2 diabetes mellitus.
