白细胞介素-2对乙型肝炎病毒基因疫苗免疫效应的影响

摘    要:目的 构建表达乙型肝炎病毒 (HBV)S蛋白的重组质粒pCR3 .1 -S作为基因疫苗 ,观察重组人白细胞介素 - 2 (rhIL - 2 )作为佐剂对其诱导BALB/c小鼠产生免疫应答的影响。方法 以不同剂量IL - 2作为佐剂 ,生理盐水作为对照 ,测定不同免疫组的血清抗HBs、淋巴细胞增殖活性、淋巴细胞杀伤活性 ,比较不同免疫组间体液和细胞免疫应答的差异。结果 抗HBsELISA滴度 ,IL - 2组与对照组比较差异无显著性 (P >0 .0 5) ;淋巴细胞增殖指数、淋巴细胞杀伤率 ,IL - 2  1 0 0 0U/ (只·次 )组、IL - 2  2 0 0 0U/ (只·次 )组与对照组比较差异有显著性 (P <0 .0 5) ,而IL - 2  50 0U/ (只·次 )组与对照组比较差异无显著性 (P >0 .0 5)。结论 一定剂量的IL - 2作为佐剂可增强HBV基因疫苗的免疫效应。

Abstract:Objective To construct recombinant plasmid of pCR3.1-S encoding protein S of hepatitis B virus as gene vaccine and investigate the effect of recombinant human interleukin 2 as an adjuvant on immune response induced by HBV gene vaccine in BALB/c mice. Methods Anti-HBs antibody to HBV major protein was measured by ELISA assay, and the activity of lymphocyte proliferation and cytotoxity was detected by 3H-TdR incorporation assay and 4-hour 51Cr release assay. Humoral and cellular immune response of different groups were then compared. Results The titers of anti-HBs antibody in serum of IL-2 group mice were not significantly higher than that of control mice(P>0.05). The difference of the activity of lymphocyte proliferation and cytotoxity in control mice and IL-2 group mice (every mouse with 500U one time) was not significant (P>0.05). However, Stimulation index and specific lysis rate of lymphocyte of IL-2 group mice (every mouse with 1000U or 2000U one time) showed significant difference compared with that of control mice (P<0.05). Conclusion Recombinant plasmid of pCR3.1-S as HBV gene vaccine could induced strong humoral and cellular immune response in BALB/c mice. IL-2 acts as an adjuvant to increase the potency of HBV gene vaccine in some certain.
