尾加压素Ⅱ对针刺预处理I/R损伤大鼠心肌细胞血管VEGF的影响

目的：研究尾加压素Ⅱ（UⅡ）在针刺抗心肌缺血再灌注损伤中的作用。方法：选择健康SD大鼠120只,随机分为6组,正常对照组（A）、模型组（B）、针刺预处理组（C）、针刺＋尾加压素低浓度组（20pmol/kg）（D）、针刺＋尾加压素高浓度组（60pmol/kg）（E）和血管紧张素Ⅱ特异性受体拮抗剂（Losartan,氯沙坦）（F）,每组20只。针刺预处理组连续3d电针预处理,第3天复制心肌缺血再灌注模型。D组、E组造模前尾静脉给药,给药后即刻造模。F组于造模前进行灌胃给药,给药后即刻造模。造模成功后常规饲养24h,处死取材,用ELISA法测定心肌组织中的VEGF。结果：心肌缺血再灌注损伤时模型大鼠心肌组织中的VEGF含量显著升高,针刺预处理、针刺结合不同剂量UⅡ以及使用UⅡ拮抗剂均可使缺血区心肌组织中的VEGF含量降低（P〈0.05）,以针刺预处理结合低剂量UⅡ效果最优（P〈0.01）。结论：UⅡ可能是抗急性心肌缺血再灌注损伤的重要作用途径之一。

Effect on VEGF in myocardial cell of acupuncture pre-conditioning I/R in jury rats on Urotensin Ⅱ

Objective：To research the effect of Urotensin Ⅱ in myocardial ischemia reperfusion injury.Methods：To select 120 healthy SD rats, divided into six groups randomly： normal group（A）,model group（B）,acupuncture precondition group （C）,acupuncture with injecting U Ⅱ at low conditions （20 pmol/kg）（D）, acupuncture with injecting U Ⅱ at high conditions （60 pmol/kg）（E）and injecting Losartan（F）,20 rats each group.Rats in C group were taken the treatment of electro-acupuncture three days continuously.In the third day, estabilished rat models of myocardial ischemia-reperfusion in the group.Rats in D, E groups were estabilished the model of myocardial ischemia-reperfusion immediately after inject U Ⅱ in caudalis vena. Rats in F group were received the administration of intragastric first, then be estabilished the model like D, E groups. Rats underwent a recovery period at least 24 hours after the model be estabilished successfully, used the clarified liquid to tast the content of VEGF by the method of ELASE.Results： The content of VEGF moved up in the condition of myocardium ischenmia-reperfusion injury, but in C, D, E and F groups the content of VEGF moved down（P〈0.05）,the effect of acupuncture with injecting U Ⅱ at low conditions is the best （P〈0.01）.Conclusion： U Ⅱ can do something in treating acute myocardial ischemia reperfusion injury.
