摘    要:目的 探讨细胞凋亡在肾小球硬化过程中的意义。方法 将48只8~10周雄性SD大鼠随机分为两组,于实验第1天、第21天对模型组分别从大鼠尾静脉注射2mg/kg阿霉素,给对照组注射等量生理盐水。第2次注药后4、12、20、28周时分别处死每组各4只大鼠,取肾行病理及电镜检测,并用末端标法凋亡情况。结果 模型组大鼠在20和28周时出现肾小铧,肾小萎缩、间质单个核细胞浸润和纤维化,肾小球、肾小雕恨细胞数显

Abstract:Objective To determine the significance of glomerular and tubular cell apoptosis in glomerulosclerosis in rats induced by adriamycin. Methods Forty eight male SD rats were randomly divided into two groups: model group and control one. The 24 rats in model group were given adriamycin(doxorubicin)2 mg/kg intravenously on the 1st day and the 21st day, respectively. At the same time, normal saline solution was given to the other 24 rats in control group by the same way. Four rats in each group were sacrificed every 8 weeks from the 4th week after the second injection. The kidneys were studied by light microscopy and electronic microscopy. Apoptotic cells in the kidney were analyzed by in situ terminal deoxynucleotidyl transferase (TdT) mediated nick end labeling. Results In rats of model group, glomerulosclerosis, tubular atrophy and interstitial mononuclear cells infiltration and fibrosis emerged, and apoptotic cells in glomeruli and tubuli significantly increased at the 20th week and the 28th week. The glomerulosclerosis index was significantly correlated to the apoptotic indexes of glomeruli and tubuli ( r s =0.831, 0.860 respectively; P <0.01 for both). Conclusion Cells elimination in glomeruli and tubuli during glomerulosclerosis in adriamycin induced nephropathy was related to apoptosis.
